摘要
为探讨高脂饲喂对五指山猪血清中TNFα、IL-1β和IL-6含量以及脂肪组织中3个基因mRNA表达水平的影响,试验将12头体重(16.44±3.08)kg的4~5月龄五指山猪随机分为2组,分别饲喂基础饲粮和含3%胆固醇的高脂饲粮12个月,然后用ELISA方法检测其血清中3种细胞因子含量,采用q RT-PCR技术对其脂肪组织中TNFα、IL-1β和IL-6 mRNA进行定量。检测结果显示高脂饲喂后肥胖组五指山猪血清中TNFα、IL-6和IL-1β水平极显著或显著高于对照组,而且在肥胖组五指山猪皮脂与腹脂组织中TNFα、IL-6和IL-1βmRNA表达量也呈现同样的变化趋势。以上结果表明高脂饲喂通过上调血清和脂肪组织TNFα、IL-6和IL-1βmRNA的表达量而激活脂肪组织慢性炎症。
The purpose of this study was to investigate TNFα, IL-1β and IL-6 levels of serum and m RNA expression levels of three genes in adipose tissue of high fat diet-induced obesity in Wuzhishan pig. Twelve pigs were randomly divided into 2 groups when mean live weight was approximately(16.44±3.08) kg. Pig were fed a corn-soybean normal protein diet(NP) or high-fat diet containing 3% cholesterol for 12 months. At the end of the trial, TNFα, IL-1β and IL-6 levels of serum was measured with enzyme-linked immunosorbent assay(ELISA), respectively SYBR Green I-based one-step real-time quantitative RT-PCR amplification was performed in quantitation of the three gene m RNA expression. The results showed that the levels of TNFα, IL-6 and IL-1β in the serum of obese group were significantly higher than those in the control group(P0.05). At the same time, these three gene m RNA expression also showed the same trend in the sebum and abdominal fat of the Wuzhishan pigs in the obese group compared with narmal group. These results indicate that high-fat diet activates adipose tissue chronic inflammation by up-regulating the expression of TNFα, IL-6 and IL-1βexpression in serum and adipose tissue.
