摘要
为了探讨Toll样受体2基因与荷斯坦牛乳腺炎抗性的相关性,本研究选取中国荷斯坦牛为试验对象构建DNA池,设计4对特异性引物,并结合克隆测序法对中国荷斯坦牛TLR_2基因编码区进行多态性筛查。结果显示,荷斯坦牛TLR_2基因编码区有6个核苷酸突变位点,分别为T602A-Intron2、G10900C-Exon2、G11047C-Exon2、A11076T-Exon2、C12077T-Exon2、T10634G-Exon2,其中T10634G-Exon2为错义突变,其余均为同义突变,SNPs突变前后等位基因频率有明显差异。6个SNPs等位基因频率分别由0.8067、0.7843、0.7959、0.7647、0.6897、0.8413突变为0.1933、0.2157、0.2041、0.2353、0.3103、0.1587。研究结果可为中国荷斯坦牛的分子抗病育种提供理论依据。
In order to investigate the correlation between the Toll like receptor-2 gene and the resistance of Holstein Cattle's mastitis, in this study, Chinese Holstein cows were selected as the experimental object to construct the DNA pooling, 4 pairs of specific primers were designed, and the TLR2 gene coding region was screened by cloned sequencing. The results showed that there were 6 nucleotide mutation sites in the TLR2 gene coding region in Holstein cattle, T602 A-Intron2, G10900 C-Exon2, G11047 C-Exon2, A11076 T-Exon2, C12077 T-Exon2 and T10634 GExon2,respectively, and T10634 G-Exon2 was a missense mutation, the rest of nucleotide mutation sites were synony mousmutations,it was shown that the alle lefre quenciesweres ignificant differences before and after SNPs mutation. The frequencies of 6 alleles of SNPs were ranged from 0.8067, 0.7843, 0.7959, 0.7647, 0.6897, 0.8413 to 0.1933, 0.2157, 0.2041, 0.2353, 0.2353, and 0.2353, respectively. The results can provide theoretical basis for molecular breeding of Chinese Holstein cattle.
