芒果苷钙盐对2型糖尿病合并非酒精性脂肪肝大鼠的影响

Effect of Mangiferin calcium salt on type 2 diabetes mellitus and nonalcoholic fatty liver in rats

摘要 目的研究芒果苷钙盐对2型糖尿病合并非酒精性脂肪肝大鼠糖脂代谢的影响。方法按照体重将雄性Wistar大鼠随机分为空白组、模型组、对照组和高、中、低剂量实验组,每组10只,除空白组外,各组均用高脂饲料配合注射链脲佐菌素复制2型糖尿病合并非酒精性脂肪肝大鼠模型。模型建立后,高、中、低3个剂量实验组分别给予480,240,120 mg·kg^(-1)芒果苷钙盐,灌胃;对照组给予125 mg·kg^(-1)二甲双胍,灌胃;空白组及模型组给予等剂量蒸馏水,每日1次,连续10周。给药结束后检测各组大鼠空腹血糖、空腹胰岛素、胰岛素抵抗指数、血清血脂含量、肝功能及血清超氧化物歧化酶活性、丙二醛、一氧化氮含量。结果空白组、模型组、对照组和高、中、低3个剂量实验组的空腹血糖含量分别为(3.21±1.20),(6.07±2.06),(4.11±1.46),(4.32±1.36),(4.46±2.13),(4.87±1.96)mmol·L^(-1),空腹胰岛素含量分别为(28.60±4.01),(33.50±3.12),(29.90±3.06),(28.20±3.17),(29.60±2.27),(31.70±3.22)m U·L^(-1),胰岛素抵抗指数分别为4.24±1.09,9.13±2.11,5.26±1.88,5.38±2.06,5.91±2.27,6.93±2.57,血清三酰甘油分别为(0.47±0.07),(0.64±0.13),(0.49±0.12),(0.49±0.10),(0.51±0.09),(0.59±0.07)mmol·L^(-1),胆固醇分别为(1.39±0.16),(1.63±0.14),(1.47±0.11),(1.40±0.17),(1.49±0.15),(1.54±0.11)mmol·L^(-1),游离脂肪酸分别为(0.62±0.16),(1.13±0.33),(0.76±0.35),(0.73±0.25),(0.81±0.28),(1.02±0.19)mmol·L^(-1),谷丙转氨酶分别为(43.6±4.2),(78.1±11.4),(60.0±9.7),(56.4±11.4),(61.6±10.3),(64.3±13.3)m U·L^(-1),血清超氧化物歧化酶活性分别为(93.2±29.3),(45.2±14.4),(70.9±20.7),(91.8±28.6),(79.9±28.7),(66.2±18.6)U·L^(-1),丙二醛分别为(4.96±1.78),(10.60±4.47),(5.82±1.44),(5.21±3.08),(5.46±2.13),(6.11±1.79)mmol·L^(-1),一氧化氮分别为(1.65±0.73),(3.16±1.27),(1.71±0.90),(1.69±0.86),(1.73±0.89),(2.04±0.54)μmol·L^(-1)。上述指标,高、中2个剂量实验组与模型组比较,差异均有统计学意义(均P<0.05)。结论芒果苷钙盐对2型糖尿病合并非酒精性脂肪肝具有一定的治疗作用。 Objective To investigate the effect of Mangiferin calcium salt on glycolipid metabolism in type 2 diabetes mellitus and nonalcoholic fatty liver rats. Methods Wistar male rats were randomly divided into blank group,model group,control group and high,middle,low doseexperimental groups. Type 2 diabetes mellitus and nonalcoholic fatty liver model rats were establish by feeding high fat food combined with traperitoneal injection of streptozotocin except blank group rats. And the high,middle and low dose groups were orally given with mangiferin calcium salt 480,240,120 mg · kg-1,the control group was orally given metformin 125 mg·kg-1,the blank and model group were given the same dose of distilled water,and 6 groups were intervened for 10 weeks. The content of fasting blood glucose( FBG),fasting insulin( FINS),insulin resistance index( IR),serum lipid content,liver function,malondialdehyde( MDA),nitric oxide( NO) and superoxide dismutase( SOD) activity were detected after administration. Results The FBG in blank group,model group,control group and high,middle,low dose experimental groups were( 3. 21 ± 1. 20),( 6. 07 ± 2. 06),( 4. 11 ± 1. 46),( 4. 32 ± 1. 36),( 4. 46 ± 2. 13),( 4. 87 ± 1. 96) mmol · L-1,the FINS were( 28. 6 ± 4. 01),( 33. 5 ± 3. 12),( 29. 9 ± 3. 06),( 28. 20 ± 3. 17),( 29. 60 ± 2. 27),( 31. 70 ± 3. 22) m U·L-1,the IR were 4. 24 ± 1. 09,9. 13 ± 2. 11,5. 26 ± 1. 88,5. 38 ± 2. 06,5. 91 ± 2. 27,6. 93 ± 2. 57, the triglyceride in serum were( 0. 47 ± 0. 07),( 0. 64 ± 0. 13),( 0. 49 ± 0. 12),( 0. 49 ± 0. 10),( 0. 51 ± 0. 09),( 0. 59 ± 0. 07) mmol·L-1,the content of total cholesterol were( 1. 39 ± 0. 16),( 1. 63 ± 0. 14),( 1. 47 ± 0. 11),( 1. 40 ± 0. 17),( 1. 49 ± 0. 15),( 1. 54 ± 0. 11) mmol·L-1,the content of free fatty acids were( 0. 62 ± 0. 16),( 1. 13 ± 0. 33),( 0. 76 ± 0. 35),( 0. 73 ± 0. 25),( 0. 81 ± 0. 28),( 1. 02 ± 0. 19) mmol · L-1,the content of alanine aminotransferase were( 43. 6 ± 4. 2),( 78. 1 ± 11. 4),( 60. 0 ± 9. 7),( 56. 4 ± 11. 4),( 61. 6 ± 10. 3),( 64. 3 ± 13. 3) m U·L-1,the activity of SOD were( 93. 2 ± 29. 3),( 45. 2 ± 14. 4),( 70. 9 ± 20. 7),( 91. 8 ± 28. 6),( 79. 9 ± 28. 7),( 66. 2 ± 18. 6) U·L-1,the content of MDA were( 4. 96 ± 1. 78),( 10. 6 ± 4. 47),( 5. 82 ± 1. 44),( 5. 21 ± 3. 08),( 5. 46 ± 2. 13),( 6. 11 ± 1. 79) mmol·L-1,the content of NO were( 1. 65 ± 0. 73),( 3. 16 ± 1. 27),( 1. 71 ± 0. 90),( 1. 69 ± 0. 86),( 1. 73 ± 0. 89),( 2. 04 ± 0. 54)μmol·L-1. Compared with model groups,the differences were statistically significant compared with middle and high dose experimental groups( P〈0. 05). Conclusion Mangiferin calcium salt has certain therapeutic effect for type 2 diabetes mellitus and nonalcoholic fatty liver.
