姜黄素对雄性小鼠生精细胞氧化损伤的保护作用 被引量:1

Protective effect of curcumin on the spermatogenic cells of oxidative damage in male mice

摘要 目的研究姜黄素对雄性小鼠生精细胞氧化损伤的影响。方法按照体重将雄性小鼠随机分为4组,每组6只:正常组、模型组和低、高2个剂量实验组。正常组注射等量0.9%Na Cl;模型组注射环磷酰胺(CTX)50 mg·kg^(-1);低、高2个剂量实验组分别灌胃给药姜黄素100,200 mg·kg^(-1),每日1次,第9天开始,腹腔注射CTX 50 mg·kg^(-1),持续10 d。第11天,小鼠脱颈椎处死。在显微镜下,用伊红染色观察精子畸形、吉姆萨染色观察精原细胞染色体畸变,用试剂盒方法测定睾丸组织的超氧化物歧化酶(SOD)活力与丙二醛(MDA)含量。结果正常组、模型组和低剂量实验组的精子畸变率分别是(6.54±1.38)%,(16.29±0.59)%,(11.97±1.64)%;这3组的精原细胞染色体畸变率分别是(4.46±1.39)%,(15.27±3.22)%,(9.95±1.62)%,模型组与正常组比较,差异有统计学意义(P<0.05);低剂量实验组与模型组比较,差异有统计学意义(P<0.05)。正常组、模型组和低、高2个剂量实验组的MDA含量分别是120.67,160.00,137.33,129.33 nmol·mg-1;这4组的SOD活力分别是373.00,321.67,337.33,345.67 U·mg-1,模型组与正常组比较,差异均有统计学意义(均P<0.05);低、高2个剂量实验组与模型组比较,差异均有统计学意义(均P<0.05)。结论姜黄素能显著降低小鼠精子畸形率、精原细胞染色体畸变率和睾丸组织的MDA含量,并能显著提高睾丸组织的SOD活力。 Objective To research whether curcumin has protective effect on the oxidative damage of spermatogenic cells of male mice.Methods Male mice were randomly divided into 4 groups,each group had 6 mouse: normal group,model group,experimental-low,-high doses groups. The model group treatment with cyclophosphamide( CTX) 50 mg·kg-1; experimental-low,-high doses groups treatment with CTX50 mg·kg-1 plus curcumin 100,200 mg·kg-1,1 time a day for 10 d.The mice were sacrificed by cervical dislocation at the time of 11 d. The sperm deformity was observed by eosin staining; the spermatogonial chromosomal aberrations was observed by Giemsa staining. The superoxide dismutase( SOD) activity and the content of malondialdehyde( MDA) in testis tissue were measured by Kit. Results The sperm aberration rate in normal group,model group and experimental-low dose group were( 6. 54 ± 1. 38) %,( 16. 29 ± 0. 59) %,( 11. 97 ± 1. 64) %,respectively;the spermatophore chromosome aberration rate in the 3 groups were( 4. 46 ± 1. 39) %,( 15. 27 ± 3. 22) %,( 9. 95 ± 1. 62) %,respectively;comparison between the model group with nomal group,the diference was significantly increased( P〈0. 05); comparison between the experimental-low dose group with model group,the diference was significantly increased( P〈0. 05).The MDA content in normal group,model group,and experimental-low,-high dose doses groups were 120. 67,160. 00,137. 33,129. 33 nmol·mg-1 respectively; the SOD activity in the 4 groups were 373. 00,321. 67,337. 33,345. 67 U·mg-1,respectively; comparison between the model group with nomal group,the difference was significantly increased( all P〈0. 05);comparison between the experimental-low dose group with model group,the difference was significantly increased( all P〈0. 05). Conclusion Curcumin can significantly decrease the content of MDA in testis,sperm malformation rate and spermatogonial cell chromosome aberration rate in mice,and can significantly increase the SOD activity of testicular tissue.
