甲酰肽受体2促进胃癌细胞侵袭及转移作用研究 被引量:2

摘要 目的通过体外胃癌细胞实验和体内裸鼠成瘤实验,探讨甲酰肽受体2(FPR2)对胃癌细胞侵袭、转移的作用。方法采用慢病毒转染技术干扰胃癌细胞SGC7901和XN0422中FPR2的表达,通过实时定量逆转录聚合酶链反应和Western blot方法检测干扰效率。利用划痕实验、Transwell实验、体内裸鼠皮下成瘤及腹腔种植实验,观察XN0422-mock、SGC7901-mock组以及XN0422-sh FPR2、SGC7901-sh FPR2组的侵袭转移、皮下成瘤和腹腔转移能力的改变。结果慢病毒感染XN0422、SGC7901细胞,FPR2表达量下降>80%。XN0422-sh FPR2组、SGC7901-sh FPR2组中侵袭、转移能力,皮下成瘤能力和腹腔转移能力均低于XN0422-mock组和SGC7901-mock组,组间比较,差异有统计学意义(P<0.05)。结论 FPR2可同时促进体外胃癌细胞的侵袭转移能力、体内皮下成瘤和腹腔转移能力。 Objective To investigate the role of the expression of formyl peptide receptor 2( FPR2) in invasion and metastasis of gastric cancer( GC) cells by using GC cells experiment in vitro and tumor formation in nude mice. Methods The expression of FPR2 in GC cells SGC7901 and XN0422 was disturbed by being transfected lentiviruses,and detected by using qRT-PCR and Western Blot. The invasion and metastasis ability,tumorigenesis and intraperitoneal metastasis of XN0422/SGC7901-mock group and XN0422/SGC7901-sh FPR2 group were compared by using wound-healing assay,transwell assay,xenografts and intraperitoneal metastasis assay.Results The expression of FPR2 in XN0422/SGC7901 cells was reduced by more than 80% infected lentivirus. In XN0422/SGC7901-sh FPR2 group,the ability of invasion and metastasis,tumorigenesis and intraperitoneal metastasis were significantly reduced comparing with XN0422/SGC7901-mock group and the difference was statistically significant( P〈 0. 05). Conclusion FPR2 promotes the ability of invasion and metastasis in GC cells in vitro as well as the ability of tumorigenesis and peritoneal metastasis in vivo.
