FUCA1基因在肝细胞肝癌中的表达及与预后的关系

The relationship between FUCA1 gene expression and invasiveness and prognosisof hepatocellular carcinoma

目的探讨肝细胞肝癌组织标本中FUCA1的表达,分析其与临床病理参数及预后的关系。方法采用Western Blot和q RT-PC技术对72例肝癌患者癌组织与癌旁的FUCA1表达水平进行检测,结合临床病理资料、随访数据,采用Kaplan-Meier和Log-rank进行预后分析,Cox回归进行危险因素的单、多因素分析。结果 FUCA1在肝癌组织学标本中较癌旁组织呈低表达,肝癌组织中FUCA1蛋白低表达与肿瘤大小、血管侵袭、卫星灶有关(P<0.05)。Kaplan-Meier生存分析和Log-rank检验指出:肝细胞肝癌中FUCA1高表达组患者比低表达组的总体生存率和无瘤生存率明显提高,差异具有显著性(P<0.05)。Cox回归预后分析发现:FUCA1表达量、肿瘤大小、血管侵袭、卫星灶以及TNM分期为评价肝癌患者预后的独立因素。结论 FUCA1在肝癌组织中的表达水平比癌旁组织明显降低,与肝癌的肿瘤大小、血管侵袭、卫星灶呈负相关且是独立的预后因子。FUCA1表达与肝癌预后联系紧密。

Objective To explore the potential role of FUCA1 gene expression in invasiveness and prognosisof hepatocellular carcinoma（HCC）. Methods A total of 72 HCC tissue samples and the corresponding adjacenttissues were examined by Western blot and q RT-PCR assays. The relationship of FUCA1 expression levels andclinicopathological variables were assessed. The Kaplan-Meier estimates and log-rank test were used to explore theoverall survival rates and disease free survival rates. The Cox regression model is used to test the multivariatesurvival analysis. All statistical tests were two-sided. Results The levels of FUCA1 in HCC was frequently down-regulated in HCC tissues compared with adjacent non-tumor tissues. The low expression of FUCA1 in HCC tissue isassociated with tumor size,tumor microsatellite formation,venous invasion status（P〈0.05）. Kaplan-Meier survivalanalysis indicated that patients with high FUCA1 level showed a positive association towards a longer survival,either overall survival（OS）or disease free survival（DFS）. Cox regression analysis found that：level of FUCA1 expression,tumor size,tumor microsatellite formation,venous invasion status and TNM stage were independent ofother prognostic factors. Conclusions FUCA1 expression is inversely correlated with tumor size,tumor microsatel-lite formation,and venous invasion status. Furthermore,low expression of FUCA1 significantly contributes to pooroutcomes of HCC,as independent prognostic factor.
