电针对食蟹猴前交叉韧带损伤后NT-3/TrkC信号通路的调节作用 被引量:2

The Regulating Role of Electroacupuncture on NT-3/TrkC Signal Pathway in Cynomolgus Monkeys after Anterior Cruciate Ligament Injury

摘要 目的观察电针对食蟹猴前交叉韧带(ACL)损伤后NT-3/Trk C信号通路的影响,探讨电针治疗ACL损伤后本体感觉减退的作用机制。方法 21只食蟹猴随机分为3组:电针干预组9只、模型对照组9只和空白对照组3只,其中对电针干预组和模型对照组进行单侧ACL损伤造模,并给予电针干预组从术后7天开始电针干预,持续时间为15 min,每天1次,共干预12周。在干预4、8、12周后,运用RT-q PCR和Western blot技术检测L2-S1脊背神经节(DRG)中NT-3、Trk C以及ACL中GAP-43的mRNA和蛋白表达水平。结果在同一时间点,与空白对照组比较,模型对照组DRG中NT-3、Trk C的mRNA、蛋白相对表达量上升,ACL中GAP-43的mRNA、蛋白相对表达量上升,差异有统计学意义(P<0.05);与模型对照组比较,电针干预组DRG中NT-3、Trk C的mRNA、蛋白相对表达量下降,ACL中GAP-43的mRNA、蛋白相对表达量上升,差异有统计学意义(P<0.05)。模型对照组NT-3、Trk C和GAP-43的mRNA和蛋白表达量随时间的增加而逐渐下降,电针干预组NT-3、Trk C和GAP-43的mRNA和蛋白表达量随时间的增加而逐渐上升,均差异有统计学意义(P<0.05)。结论电针能够通过激活NT-3/Trk C信号通路,提升DRG中NT-3、Trk C和局部GAP-43的表达量,在ACL本体感觉的康复中发挥重要的作用。 Objective To observe the effect of electroacupuncture( EA) on NT-3/Trk C signal pathway after anterior cruciate ligament( ACL) injury of cynomolgus monkeys,and to study its mechanism for treating proprioception loss after ACL injury. Methods Totally 21 cynomolgus monkeys were randomly divided into 3 groups,the EA intervention group(n = 9),the model control group(n = 9),and the blank control group(n = 3). The model control of unilateral ACL injury was performed in the EA intervention group and the model control group. EA was performed to monkeys of the EA intervention group from the 7 th day after operation,1 time per day,lasting 15 min,12 weeks in total. At week 4,8,12 after intervention,mRNA and protein expressions of NT-3 and Trk C in L2-S1 dorsal root ganglion( DRG) and GAP-43 in ACL were detectedby RT-q PCR and Western blot. Results Compared with the blank control group,mRNA and protein expressions of NT-3 and Trk C in DRG decreased,mRNA and protein expressions of GAP-43 in ACL increased in the model control group at the same time points(P〈0. 05). Compared with the model control group,mRNA and protein expressions of NT-3 and Trk C in DRG increased,mRNA and protein expressions of GAP-43 in ACL increased in the EA intervention group(P〈0. 05). In the model control group,mRNA and protein expressions of NT-3,Trk C,and GAP-43 decreased gradually as time went by. In the EA intervention group,mRNA and protein expressions of NT-3,Trk C,and GAP-43 increased gradually as time went by(P〈0. 05). Conclusions NT-3/Trk C signal pathway could be activated by EA. The expressions of NT-3 and Trk C in DRG,and the expression of local GAP-43 were also elevated by EA. EA played an important role in the rehabilitation of ACL proprioception.
