摘要
目的通过单因素试验获得牛肝菌膳食纤维的提取工艺,并对其干预便秘的作用进行研究。方法以提取温度、料液比、提取时间及提取次数等因素对牛肝菌膳食纤维的提取工艺进行优化;将牛肝菌粗粉按100、200、400mg·kg-1作用于健康ICR小鼠,用复方地芬诺酯造便秘模型,检测小肠推进率。结果牛肝菌膳食纤维的提取工艺是提取温度80℃,料液比1∶30,提取时间1次2h,提取次数3次;跟模型组相比,牛肝菌高剂量组对便秘具有干预作用,且作用与阳性药组基本相当。结论牛肝菌膳食纤维水提工艺稳定可靠,糖得率高,且牛肝菌具有良好的便秘干预作用。
OBJECTIVE The extraction technology of dietary fiber from Boletus edulis was obtained by single factor test,and the effect of its intervention on constipation was studied. METHODS The extraction process of Boletus edulis dietary fibers( BEDF) was optimized by extracting temperature,ratio of material to liquid,extraction time and extraction times. Healthy ICR mice were treated with crude powder of Boletus edulis at the dosage of 100,200,400 mg·kg^-1,then constipation model was established by compound Diphenoxylate,and propulsion rate of small intestine was tested to evaluate the intervention of boletus edulis against constipation. RESULTS The optimum extraction conditions of BEDF were extracted at 80℃,ratio of liquid to material in 1 ∶ 30,2 hours per time and for 3 times. Compared with the model group,high dose group of boletus edulis has an intervention effect on constipation,and the effect is almost similar to the positive drug group. CONCLUSION BEDF optimized by single-factor test is stable and reliable,the yield of sugar is high. BEDF has good constipation intervention,and its specific mechanism needs further study.
