摘要
目的:探讨左归丸降低肾阴虚大鼠血清cAMP/cGMP比值与抗氧化作用的关联性。方法:48只SD大鼠随机分为正常组、模型组及左归丸低、高剂量组,除正常组外,其余各组大鼠每天灌胃甲状腺素(T4)150mg/kg及利血平1mg/kg,复制肾阴虚模型,于造模同时正常组及模型组每天灌胃0.9%氯化钠溶液,左归丸低、高剂量组每天灌胃左归丸水煎液136.5、273g/kg,连续给药7d,运用酶联免疫法检测血清环磷酸腺苷(cAMP)、环磷酸鸟苷(cGMP)、T4水平,同时检测血清肌酐(Cre)水平、血清及肾脏超氧化物歧化酶(SOD)活性、丙二醛(MDA)水平,并对血清SOD、cAMP、cGMP、cAMP/cGMP比值与各项指标进行相关性分析。结果:与模型组比较,左归丸低、高剂量均能降低肾阴虚大鼠血清cAMP水平、血清cAMP/cGMP比值、血清Cre及肾脏MDA水平(P<0.05,P<0.01),升高大鼠血清SOD活性(P<0.01);左归丸高剂量能升高大鼠血清cGMP水平(P<0.05);Pearson相关性分析结果显示肾阴虚大鼠血清cAMP与血清SOD活性呈负相关,与肾脏SOD及血清Cre呈正相关;血清cGMP与血清MDA呈负相关;血清cAMP/cGMP比值与血清SOD活性呈负相关,与血清MDA、Cre水平呈正相关;血清SOD与血清Cre呈负相关。结论:左归丸可能通过提高血清SOD活性,增强机体抗氧化能力,进而降低肾阴虚大鼠血清cAMP水平、cAMP/cGMP比值及Cre水平,改善肾阴虚症状。
Objective: To study the correlation between the ratio of cAMP/cGMP in serum and anti-oxidation of Zuogui Pills in kidney-yin deficiency model rats. Methods: Forty-eight SD rats were randomly divided into normal group, model group, high-dose and low-dose group of Zuogui Pills. Except for normal group, other rats were intragastrically administrated with tetraiodothyronine(T4) 150 mg·kg^-1·d^-1 and Reserpine 1 mg·kg^-1·d^-1 to induce kidney-yin deficiency model; meanwhile, rats in normal group and model group were intragastrically administrated with saline solution, in low-dose and high-dose group of Zuogui Pills were intragastrically administrated with Zuogui Pills 136.5 g·kg^-1·d^-1, 273 g·kg^-1·d^-1, for 7 days, respectively. The levels of cyclic adenosinemonophosphate(cAMP), cyclic guanosine monophosphate(cGMP) and T4 in serum were detected using the enzyme-linked immunosorbent assay. In addition, the levels of creatinine(Cre) in serum, malondialdehyde(MDA) and SOD in serum and kidney were detected. Pearson relational analysis was used to analyze the associations of the levels of SOD, cAMP, cGMP and the ratio of cAMP/cGMP in serum with other biochemical indicators. Results: Compared with the model group, the levels of cAMP, Cre, the ratio of cAMP/cGMP in serum and MDA in kidney were significantly decreased(P〈0.05, P〈0.01), the activity of SOD in serum was significantly increased in high-dose and low-dose group of Zuogui Pills(P〈0.01). And the highdose of Zuogui Pills could improve the levels of cGMP in serum(P〈0.05). Pearson correlation analysis showed that there were negative correlations between cAMP and SOD in serum, cGMP and MDA in serum, the ratio of cAMP/cGMP and SOD in serum, SOD and Cre in serum, and positive correlation between cAMP and Cre in serum, cAMP in serum and SOD in kidney, the ratio of cAMP/cGMP and MDA in serum, the ratio of cAMP/cGMP and Cre in serum. Conclusion: The mechanisms of Zuogui Pills on kidney-yin deficiency may association with increasing the activity of SOD in serum, enhancing the antioxidant capacity of the body, and then decreasing the levels of cAMP, Cre and the ratio of cAMP/cGMP.
