HPLC法同时测定藏药斑花黄堇中3种生物碱的含量 被引量:2

摘要 目的:采用HPLC法同时测定藏药斑花黄堇中异紫堇定、比枯枯灵、四氢巴马汀3种生物碱的含量。方法:采用Eclipse XD8-C_(18)柱(250mm×4.6mm,5μm);流动相甲醇(A)-0.1mol/L醋酸铵(0.01%磷酸,调pH为6.0)溶液(B),梯度洗脱:0~5min,40%A;5~10min,40%~49%A;10~30min,49%A,流速1.0mL/min,检测波长290nm,柱温35℃。结果:3种生物碱成分达到了良好的分离,R>1.5;在线性范围内的线性范围良好,r≥0.9998,平均回收率在99.97%~100.04%,R S D为0.17%~0.42%(n=5)。结论:所使用的方法快捷、灵敏度高、稳定性好,可用于藏药斑花黄堇3种生物碱成分的含量测定,同时为藏药材斑花黄堇的质量控制研究提供了一定的科学依据。 Objective: To detect 3 kinds of alkaloids(isocorydine, bcuculline, tetrahydropalmatine) in Corydalis conspersa Mazim. by high performance liquid chromatography(HPLC) method at the same time. Methods: Eclipse XD8-C18 column(250 mm×4.6 mm, 5μm) was used. The moving phase was composed of methanol(A) and 0.1 mol/L ammonium acetate(0.01%phosphoric acid, p H=6.0)(B). Linear gradient mode was as follows: 0-5 min, 40%A; 5-10 min, 40%-49%A; 10-30 min, 49%A. The flow rate of the mobile phase was set at 1.0 m L/min, the detector wavelength was 290 nm and the column temperature was 35℃. Results: Three kinds of alkaloids showed good separation and satisfactory linearity(r≥0.9998) within the concentration ranges tested. The average recovery rate was in the range of 99.97%-100.04%. with the RSD between 0.17%-0.42%(n=5). Conclusion: The method used is rapid with high sensitivity and good stability. It can be used for simultaneous determination of the contents of 3 kinds of alkaloids in Corydalis conspersa Mazim., at the same time, providing a scientific basis for the quality control of Corydalis conspersa Mazim..
