青钱柳叶对2型糖尿病大鼠糖脂代谢影响 被引量:19

Effects of Cyclocarya paliurus on glucose and lipid metabolism in T2D rats

摘要 目的:通过研究青钱柳叶水提取物(CPE)对2型糖尿病(T2D)大鼠的糖脂代谢的作用,探讨青钱柳叶的"甘寒"药性。方法:雄性SD大鼠,随机分为空白组10只和造模组90只,其中空白组给予普通饲料喂养,造模组给予高脂饲料喂养8周。第9周,造模组以30mg/kg一次性腹腔注射链脲佐菌素(STZ),并于72h后选择空腹血糖>11.1mmol/L的成模大鼠随机分为模型组、二甲双胍组、CPE低、中、高剂量组,每组10只;空白组和模型组给予0.9%氯化钠溶液,二甲双胍组给予二甲双胍混悬液0.3g/kg,CPE低、中、高剂量组分别给予青钱柳叶水提取物0.18、0.36、1.08g/kg。连续给药4周,检测糖脂、环磷酸腺苷、内分泌免疫等代谢指标。结果:与空白组比较,造模组体质量显著下降(P<0.01);FBG、TC、TG、LDL、c AMP、c GMP、PPARγ、T3、T4、IL-6、TNF-α、TP、ALB、UA显著升高(P<0.05,P<0.01),HDL显著下降(P<0.05);与模型组比较,CPE低、中、高剂量组FBG、TC、TG、c AMP、T3、T4、TNF-α、TP、UA显著下降(P<0.05);CPE中、高剂量组血清中PPARγ、IL-6、ALB含量显著减低(P<0.05,P<0.01),HDL显著升高(P<0.05)。结论:CPE调节阴虚内热型T2D大鼠糖脂代谢和环核苷酸代谢紊乱、抑制内分泌免疫功能亢进,其机制可能与青钱柳叶具有"甘寒"药性有关。 Objective: To investigate the effects of Cyclocarya paliurus Leaves extract(CPE) on glucose and lipid metabolism in type 2 diabetes mellitus(T2 D) rats and to explore its properties of ‘Sweet and Cold'. Methods: Male SD rats were randomly divided into blank group(n=10) and model group(n=90). The blank group was given normal feed; the model group was fed with high fat feed for 8 weeks. At the 9 th week, STZ was injected intraperitoneally into the model group at a dose of 30 mg/kg. After 72 hours, the model rats were randomly divided into model group, metformin group, CPE low, medium and high dose group, 10 rats in each group. The blank group and model group were given 0.9% sodium chloride solution, metformin group was given metformin suspension 0.3 g/kg, CPE low, medium and high dose group were given Cyclocarya paliurus leaves extract 0.18, 0.36, 1.08 g/kg. Continuous administration for 4 weeks, detection of glycolipid, cyclic AMP, endocrine and other metabolic indicators. Results: Compared with the blank group, the body weight of the model group decreased significantly(P〈0.01). The FBG, TC, TG, LDL, c AMP, c GMP, PPARγ, T3, T4, IL-6, TNF-α, TP, ALB, UA were increased(P〈0.05, P〈0.01), HDL decreased significantly(P〈0.05). Compared with the model group, the levels of FBG, TC, TG, c AMP, T3, T4, TNF-α, TP and UA in the CPE low, medium and high dose groups were significantly decreased(P〈0.05, P〈0.01). The contents of PPARγ, IL-6 and ALB were decreased, HDL increased significantly in the medium and high group(P〈0.05, P〈0.01). Conclusion: CPE regulates the glucose and lipid metabolism, attenuate cyclic nucleotide metabolism disorder and inhibits the endocrine immune function hyperactivity in T2 D rats with yin deficiency and dryness-heat. The mechanism may be is related to the properties of ‘Sweet and Cold'.
