党参提取物抗运动性疲劳机制研究 被引量:8

摘要 目的:研究中药党参提取物抗运动性疲劳的效应及机制.方法:随机将昆明小鼠分为空白对照组、党参提取物低剂量组(100mg/(kg·d))、中剂量组(300mg/(kg·d))、高剂量组(600mg/(kg·d)).喂养20d后,检测其负重力竭游泳时间、肝糖原含量、运动后血乳酸、血清尿素氮含量以及腓肠肌PGC-1α蛋白的表达.测量结果:党参提取物能够显著增加昆明小鼠力竭游泳时间,并显著降低血乳酸含量(p<0.05);此外高剂量组中血清尿素氮含量降低了28.64%(p<0.01),肝糖原含量升高了30.30%(p<0.01);Western Blot检测结果显示,高、中、低剂量组PGC-1α表达较空白组均有显著提高(p<0.05).结论:说明党参提取物很可能是通过提高PGC-1α蛋白的表达来减少代谢产物的堆积,增加糖原含量,从而达到抗运动性疲劳的作用. Objective:To study the effect and mechanism of Codonopsis pilosula extract against exercise-induced fatigue.Methods:Kunming mice were randomly divided into blank control group,low dose group(100 mg/(kg·d)),medium dose group(300 mg/(kg·d))and high dose group(600 mg/(kg·d)).After feeding 20 d,their exhaustive swimming time,liver glycogen content,blood lactic acid content,serum urea nitrogen and the expression of peroxisome proliferator-activated receptor gamma coactivator 1-alpha(PGC-1α)protein in the gastrocnastus were measured.The results showed that the extract of C.pilosula significantly increased the exhaustive swimming time and significantly decreased the content of blood lactate in Kunming mice(p〈0.05).In addition,the content of serum urea nitrogen in the high dose group decreased by 28.64%(p〈0.01).The content of PGC-1αincreased by 30.30%(p〈0.01).The results of Western Blot showed that the expression of PGC-1αin the high,medium and low dose groups significantly increased compared with that of the blank group(p〈0.05).Conclusion:C.pilosula extract may play a role in resistance to exercise-induced fatigue by increasing the expression of PGC-1αprotein,reducing the accumulation of metabolites and increasing the content of glycogen.
