摘要
目的探讨血管内皮生长因子(VEGF)转染内皮祖细胞(EPCs)移植对勃起功能障碍(ED)大鼠阴茎勃起的影响。方法8周龄Wistar雄性大鼠通过喂养高脂饲料建成ED动物模型24只,随机分为3组:Ⅰ组为对照组、Ⅱ组为EPCs移植组、Ⅲ组为YEGF基因转染EPCs移植组。分别给予EPCs细胞悬液及VEGF基因转染的EPCs悬液海绵体移植,对照组给予注射等量生理盐水,28d后进行药物勃起实验,新生毛细血管计数、反转录-聚合酶链反应(RT-PCR)及酶联免疫吸附试验(ELISA)法检测VEGF及细胞间黏附分子-1(ICAM-1)。结果Ⅲ组ICAM-1浓度为(1090.44±158.38)pg/ml明显低于Ⅱ组[(1654.27±232.14)pg/ml,P=0.045],Ⅱ组明显低于Ⅰ组的(3860.54±456.18)pg/ml,P=0.015);Ⅲ组VEGF表达为(768.13±134.37)pg/ml高于Ⅱ组[(579.79±68.62)pg/ml,P=0.025],Ⅱ组VEGF表达高于Ⅰ组[(54.45±19.37)pg/ml,P=0.048];Ⅲ组大鼠勃起次数(2.30±0.67)大于Ⅱ组(1.83±0.879,P=0.033);Ⅲ组新生血管数量达(143.32±49.27)明显高于Ⅱ组(73.69±23.32,P=0.001),Ⅱ组新生血管数量明显大于Ⅰ组(46.31±11.17,P=0.024)。结论VEGF基因转染EPCs移植可上调ED大鼠阴茎海绵体VEGF表达、促进海绵体毛细血管再生,改善阴茎血管内皮功能及阴茎勃起功能。
Objective To investigate the the effect of vascular endothelial growth factor (VEGF) gene transfected endothelial progenitor cell (EPCs) transplantation on penile for erectile dysfunction (ED) mice. Methods 24 male Wistar mice, 8 weeks, were made from animal models for ED by feeding high fat forage. They were randomly divided into 3 groups: group Ⅰ (control), group Ⅱ (EPCs transplantation) and group Ⅲ (VEGF gene transfection EPCs transplantation). EPCs and VEGF gene transfection EPCs were administered for group Ⅱ and group m respectively. Saline was given to group Ⅰ. Drug erectile experiment, newly born capillaries, ICAM - 1 and VEGF detection was performed in 28 days. Results The expression of ICAM - 1 in corpus cavernosum penis in group Ⅲ was ( 1 090.44 ± 158.38) pg/ml, which was lower than that in group Ⅱ [ ( 1 654. 27 ± 232. 14) pg/ml, P = 0. 0451. And which of group Ⅱ was lower than that of group Ⅰ [ (3 860. 54 ±456. 18) pg/ml, P =0. 015]. The expression of VEGF in corpus cavernosum penis in group Ⅲ was (768.13 ± 134.37) pg/ml, it was higher than that in group Ⅱ [(579.79± 68.62) pg/ml, P= 0.0251, and it was higher than that of group I [(54.45± 19. 37) pg/ml , P = 0. 048 ]. The erectile times in group m was 2 30 ± 0 67 and greater than which of group Ⅱ ( 1.83 ± 0. 87, P =0. 033 ). The number of newly born capillaries in group Ⅲ was 143.32 ± 49. 27 and more than which of group Ⅱ (73.69 ± 23.32, P = 0. 001 ). The number of newly born capillaries in group Ⅱ was bigger than which of group Ⅰ (46. 31 ± 11.17, P = 0. 024 ). Conclusion VEGF gene transfection endothelial progenitor cell transplantation can promote the expression of VEGF in orpus cavernosum penis, regeneration of newly born capillaries, vascular endothelial function and erectile function for ED mice.
