摘要
为进一步探讨前期报道的新型人雌激素受体ERα30的功能及其作用机制,本研究应用EdU法研究新型人雌激素受体ERα30过表达对人乳腺癌细胞MDA-MB-231增殖能力的影响,并通过实时荧光定量RT-PCR检测ERα相关靶基因的差异表达。结果显示,ERα30过表达促进了MDA-MB-231细胞的增殖,同时上调了c-jun及EGFR基因的表达,提示ERα30可能通过非经典机制发挥作用。本研究为后续深入研究ERα30在乳腺癌发生发展中的作用提供了良好的基础,为乳腺癌的诊断、治疗及预后提供新的视角。
To investigate the function and mechanism of novel human estrogen receptor ERa30 that early reported, EdU assay was performed in this research to detect the effect on proliferation of ERα30 over-expression in human breast cancer cell MDA-MB-231, and the differential gene expression of ERα associated target gene was tested by Real time RT-PCR. The result showed that over-expression of ERα30 in MDA-MB-231 cell accelerated cell proliferation, and up-regulated the expression of c-jun and EGFR gene, indicating ERα30 may mediate the "nonclassical" pathways. This study lays a good foundation for further researches about the role of ERα30 in the development of breast cancer, which provides new perspective for diagnosis, treatment, and prognosis of breast cancer.
