摘要
本研究采用免疫组化法检测42例乳腺癌组织及相应的42例癌旁组织中PD-L1的表达,并分析PD-L1表达与乳腺癌临床病理特征的关系;采用ELISA方法测定这42乳腺癌患者手术前血清PD-L1的表达及另外18正常对照血清中PD-L1的表达,并分析PD-L1在血清中表达与乳腺癌组织中的表达关联程度及其临床意义。研究结果表明:乳腺癌患肿瘤组织中PD-L1表达高于癌旁组织(p〈0.05),PD-L1高表达与肿瘤大小、肿瘤分化、淋巴结转移、远处转移相关(p〈0.05),而与年龄无关(p〉0.05)。进一步的分析显示:乳腺癌患者血清PD-L1表达显著高于正常人(p〈0.001),且乳腺癌患者血清PD-L1的表达与癌组织中PD-L1的表达显著相关(r=0.811,p〈0.001)。PD-L1在乳腺癌组织及血清中均高表达,两者显著相关,高表达PD-L1与乳腺癌进展及预后不佳相关。我们的研究初步表明采用免疫组化及ELISA检测乳腺癌肿瘤组织及外周血中PD-L1的表达,对于解释PD-L1与乳腺癌发生、发展及预后的关系,对乳腺癌的诊治及临床应用具有意义。
In this study, the expression of PD-L1 in 42 cases of breast cancer and 42 cases of adjacent tissues was detected by immunohistochemistry, and the relationship between PD-L1 expression and clinicopathological features of breast cancer was analyzed. ELISA was used to detect the expression of PD-L1 in the serum of 42 preoperative breast cancer patients and the serum of 18 normal people, and the relationship between the expression of PD-L1 in serum and in breast cancer tissues and its clinical significance were analyzed. The results showed that the expression of PD-L1 in breast cancer tissues was higher than that in adjacent tissues (p〈0.05), and the high expression of PD-LI was related with tumor size, tumor differentiation, lymph node metastasis and distant metastasis (p〈0.05), but it had no relationship with age (p〉0.05). Further analysis indicated that the expression of PD-L1 in the serum of breast cancer patients was significantly higher than that in the normal people (p〈0.001), moreover, it was closely related with the expression of PD-L1 in cancer tissues (r=0.811, p〈0.001). PD-L1 had high expression both in breast cancer tissues and serum, and the two were significantly related, and PD-L1 with high expression was associated with breast cancer progression and poor prognosis. Our study preliminarily showed that the expression of PD-L1 in breast cancer tissue and peripheral blood detected by immunohistochemistry and ELISA was useful in the explanation of the relationship between PD-L1 and the occurrence, development and prognosis of breast cancer, and it was significant for the diagnosis and treatment of breast cancer and its clinical application.
