半夏泻心汤对脾虚便秘小鼠肠道菌群与肠黏膜的影响 被引量:20

Effect of Banxia Xiexin Decoction (半夏泻心汤) on Intestinal Flora and Intestinal Mucosa in Mice with Spleen-deficiency Constipation

摘要 目的:观察半夏泻心汤对脾虚便秘小鼠肠道菌群与肠黏膜的影响。方法:将24只昆明种小鼠随机分为正常组、模型组、枳术汤组和半夏泻心汤组,每组6只。模型组、枳术汤组、半夏泻心汤组采用灌胃番泻叶水煎液,7 d后控制饮食,饥饱失常8 d,构建小鼠脾虚便秘模型。造模结束后,半夏泻心汤组、枳术汤组分别采用半夏泻心汤和枳术汤灌胃,模型组和正常组均采用蒸馏水灌胃,共6 d,观察小鼠的肠道菌群变化及小肠病理切片情况。结果:半夏泻心汤组、枳术汤组、模型组所有肠道菌群数量均明显多于正常组(P<0.05),半夏泻心汤组和枳术汤组所有肠道菌群数量均少于模型组(P<0.05),半夏泻心汤组的双歧杆菌数量明显少于枳术汤组(P<0.05);半夏泻心汤组的乳酸杆菌、大肠杆菌、肠球菌少于枳术汤组,但差异均无统计学意义(P>0.05)。模型组肠道发生炎症反应及杯状细胞明显减少。枳术汤组空肠组织绒毛形状由短锥形变成指尖形,盲肠组织中的杯状细胞未见明显恢复或增多,隐窝深度也未见明显变浅;半夏泻心汤回肠组织的炎症症状改善明显,小肠绒毛完整性恢复,固有层排列十分紧密有序,杯状细胞数量明显恢复,隐窝深度变浅。结论:半夏泻心汤能有效调节脾虚便秘小鼠紊乱的肠道菌群,改善炎症症状,恢复小肠绒毛的完整性,使黏膜隐窝深度变浅。 Objective: To observe the effect of Banxia Xiexin decoction (半夏泻心汤, BXXXD) on intestinal flora and intestinal mucosa in mice with spleen-deficiency constipation. Methods: The 24 Kumning mice were randomly and equally divided into normal group, model group, Zhizhu decoction (枳术汤) group, and BXXXD group. The mice in model group, Zhizhu decoction group, and BXXXD group were given decoction of senna by garage; 7 days later, these mice were given an irregular diet for 8 days to establish a mouse model of spleendeficiency constipation. After model establishment, the BXXXD group and Zhizhu decoction group were given BXXXD and Zhizhu decoction respectively by gavage, and the model group and normal group were given dis- tilled water by gavage; the course of treatment was 6 days for all groups. The pathological sections of the small intestine were examined after treatment. Results: The number of intestinal flora in BXXXD group, Zhizhu decoction group and model group were significantly higher than those of normal group (P〈0.05); The number of intestinal flora in BXXXD group and Zhizhu decoction group was less than that of model group (P〈0.05); The number of bifidobacteria in BXXXD group was significantly less than that of Zhizhu decoction group (P〈0.05). Lactobacillus, Escherichia coli and enterococci in BXXXD group were less than those of Zhizhu decoction group, but the difference was not statistically significant (P〉0.05). Intestinal inflammatory response and a significant reduction in goblet cells were seen in the model mice with spleen-deficiency constipation. After treatment with Zhizhu decoction, the jejunal tissue had fingertip-shaped villi instead of short cone-shaped villi, the cecal tissue showed no significant increase in goblet cells, and the depth of intestinal crypts was not significantly decreased. After treatment with BXXXD, the ileal tissue showed a significant relief of inflammatory symptoms, the integrity of small intestinal villi was restored, the lamina propria was in tight and ordered arrangement, the number of goblet cells was significantly increased, and the depth of intestinal crypts was decreased. Conclusion: BXXXD can relieve inflammatory symptoms, restore the integrity of small intestinal villi, and decrease the depth of intestinal crypts.
