石蜡切片中卵巢癌基因1第2外显子PCR-SSCP检测法建立 被引量:1

摘要 目的建立从石蜡切片中检测卵巢癌基因(OVCA)1第2外显子的聚合酶链反应(PCR)-单链构象多态性(SSCP)检测方法。方法从石蜡切片中提取基因组DNA;以基因组DNA为模板,PCR扩增OVCA1第2外显子;非变性聚丙烯酰胺凝胶电泳检测PCR产物SSCP,银染显色。结果 PCR扩增片段经琼脂糖凝胶电泳检测可见条带位置正确,条带单一。对PCR扩增片段进行SSCP检测,可见清晰条带。结论成功建立了可用于石蜡切片中OVCA1第2外显子的PCR-SSCP检测方法。
