摘要
目的:观察补正续骨丸对大鼠的急性毒性。方法:因采用经口灌胃途径,无法测定出补正续骨丸的半数致死量(LD50),所以,将补正续骨丸配置浓度调整为能完成经口灌胃操作的最大浓度,灌胃量为大鼠所能承受的最大剂量,进行急性毒性观察。结果:该药物采用经口灌胃途径给药的最大浓度为0.56 g/mL,大鼠按照体质量最大的给药剂量为2.5 mL/100 g,2次/d给药,最终给药剂量为:27.8 g/kg,该剂量为成人(60 kg体质量)每日口服剂量的83.4倍,实验过程中,大鼠未出现死亡情况,连续给药2周,病理结果显示,肝、心、脾、肺、肾脏未见病理改变。结论:补正续骨丸以27.8 g/(kg/d)体质量的剂量给予大鼠经口灌胃,连续给药2周,未见毒性反应。
Objective: To observe the acute toxicity of Buzheng Xugu Pill in mice. Method: The way of gavaging through mouth could not measure the Half Lethal Dose( LD50) of Buzheng Xugu Pill,so we adjusted the concentration to the maximum one,and the gastric volume was the maximum dose which the mice can bear. Result: The maximum concentration by the way of gavaging through mouth was 0. 56 g/mL,and the maximum dose was 2. 5 mL/100 g based on weight for twice a day. The final dosage of administration was 27. 8 g/kg,which was 83. 4 times of adult( 60 kg) for one day.The mice did not die during the test,and the pathology results of liver,heart,spleen,lung and kidney did not have changes within 2 weeks. Conclusion: By the way of gavaging through mouth of Buzheng Xugu Pill with the dose of 27. 8 g/( kg·d) during 2 weeks,the test did not appear toxic reaction.
