摘要
目的探索辛伐他汀对大肠癌细胞系HCT116组织因子(TF)表达的影响。方法用实时荧光定量PCR和流式细胞术检测给药前后大肠癌细胞系HCT116的TF m RNA和TF膜蛋白表达含量,并评价其表达水平差异。结果PCR结果显示,相对于空白组,10^(-7)mol/L和10^(-5)mol/L辛伐他汀浓度组TF m RNA的表达倍数表分别为0.693±0.052和0.280±0.021,差异有统计学意义(P<0.05和P<0.01)。流式细胞术的检测结果表明,与空白组(26.25±0.639)比较,10^(-7)mol/L和10^(-5)mol/L辛伐他汀浓度组TF膜蛋白表达的数学平均数分别为19.76±0.550和15.45±0.518,差异非常显著(P均<0.01)。结论辛伐他汀可剂量依赖地下调HCT 116细胞的TF m RNA和TF膜蛋白的表达。
ObjectiveTo study the effect of simvastatin on the expression of tissue factor(TF)in colorectal cancer cell line HCT116.MethodsThe expression of TF m RNA and membrane protein in colorectal cancer cell line HCT116 before and after administration of simvastatin was detected by real-time fluorescence quantitative PCR and flow cytometry,respectively.ResultsThe results of PCR showed that,as compared to the blank group,the relative folds of TF m RNA in 10^-7mol/L and 10^-5mol/L simvastatin group was 0.693±0.052 and 0.280±0.021,respectively,and the difference was statistically significant(P0.05and P〈0.01).The results of flow cytometry showed that,the mathematical mean of TF membrane protein expression in 10^-7mol/Land 10^-5mol/L simvastatin group was 19.76±0.550 and 15.45±0.518,respectively.As compared with the blank group(26.25±0.639),TF expression values decreased significantly(all P〈0.01).ConclusionSimvastatin can dose-dependently down-regulate the expression of TF m RNA and membrane protein in HCT116 cells.
