小鼠尾静脉与内眦静脉丛回输血小板方法的比较

摘要 目的比较小鼠尾静脉与内眦静脉丛回输血小板的操作难度及回输效果。方法体外分离血小板(10×108/m L),分别采用尾静脉和内眦静脉丛注射法将等量的calcein标记的血小板回输到8周C57 BL/6小鼠体内,比较两种方法的操作时间和成功率;对注射血小板的小鼠立即从眼眶采血,通过流式细胞仪和荧光显微镜检测回输到小鼠体内的血小板数;对采用两种不同方法成功回输等量血小板的小鼠24 h采血一次,通过流式细胞仪和荧光显微镜检测回输到小鼠体内的血小板的生存周期。结果经尾静脉和内眦静脉丛回输等量血小板:(1)平均用时分别为94 s和45.3s,成功率分别为40%和100%(P<0.05);(2)在0 h,被标记的血小板数占总血小板数的百分比分别为12.38%和12.35%,两组间差异无统计学意义(P>0.05);(3)在48 h、96 h,回输进体内的血小板分别下降44%、64%和88%、89%,两组间差异无统计学意义(P>0.05)。结论两种注射法回输到小鼠体内的血小板数没有差异,且不影响血小板的生存周期。小鼠眼内眦静脉丛回输血小板,操作更简单,成功率更高。 Objective Compare the operation difficulty and the effect of mouse tail vein and inner canthus venous plexus on platelet transfusion. Methods Lsolate platelets( 10 × 10^8/m L) in vitro were injected into the 8 weeks C57 BL/6 mouse from the tail vein and the inner canthus vein,and compare the operation time and success rate of the two methods; Mouse injected with platelets were drawn blood from another eye immediately,and the number of platelets returned to the mouse was measured by flow cytometry and fluorescence microscopy. The mouse that had been successfully returned the same amount of platelets in two different methods were drawn blood onceevery 24 h,and the platelet survival cycle in mouse was detected by flow cytometry and fluorescence microscopy. Results When returned the same amount of platelets from the tail vein and the inner canthus vein: 1. the average used time was 94 s and 45. 3 s,the success rates was 40%and 100%( P〈0. 05). 2. at 0 h,the percentage of labed platelets count to total platelets count was 12. 38%and 12. 35% respectively,and there was no significant statistical difference between two groups( P〉0. 05). 3.at 48 h、96 h,the number of the platelets that returned to mouse were decreased by 44% and 64%,88% and89%,respectively,And there was no significant statistical difference between two groups( P〈0. 05).Conclusion There was no difference in the number of injected platelets in mouse by two methods,and the survival time of injected platelets was not affected. It is easier and more successful to inject platelets through the mouse inner canthus veniplex.
