摘要
目的:研究杜仲叶提取物以及其活性成分绿原酸对体外结肠癌侵袭与迁移的影响。方法:采用HPLC法检测了杜仲叶提取物中绿原酸的含量;采用了Transwell小室法检测了杜仲叶提取物及绿原酸对结肠癌细胞株HCT116、LOVO侵袭迁移能力的影响。结果:杜仲叶提取物处理过的HCT116细胞相对于空白样品侵袭减少49.72%;绿原酸处理过的HCT116细胞相对于空白样品侵袭相对减少66.32%。杜仲叶提取物处理过的LOVO细胞相对于空白样品侵袭相对减少37.16%;绿原酸处理过的LOVO细胞相对于空白样品侵袭相对减少50.11%。迁移实验也遵循同样的规律。结论:杜仲叶提取物具有抑制结肠癌细胞侵袭和迁移的能力,绿原酸的效果更为显著。绿原酸可能是杜仲叶这种生物活性的关键成分。
Objective: The study was aimed to observe the impact of Eucommia ulmoides leaves extract and chlorogenic acid on invasion and migration of colon cancer cells. Methods: HPLC was applied to determine chlorogenic acid and transwell chambers method was applied to detect inhibition rate on invasion and migration of HCT116,Lo Vo cell line. Results: Invasion ratio of HCT116 cells previously treated with Eucommia ulmoides leaves extract solution reduced 49. 72% relative to that of the blank group,and that of chlorogenic acid was 66. 32%. Invasion reduce of ratio of Lo Vo cells previously treated with Eucommia ulmoides leaves extract solution reduced 37. 16% relative to the blank group,and that of chlorogenic acid was 50. 11%. Migration ratio followed the same routine. Conclusion: These results suggested that Eucommia ulmoides leaves extract could inhibit invasion and migration of colon cancer cells. Chlorogenic acid may be a key ingredient of the biological activity.
