热敏灸对肠易激综合征模型大鼠下丘脑CRH、CRF及肾上腺GR的影响 被引量:5

Effect of Heat-Sensitive Moxibustion on CRH and CRF in Hypothalamus and GR in Adrenal Gland of IBS Rats

摘要 目的:观察热敏灸对肠易激综合征(irritable bowel syndrome,IBS)大鼠下丘脑促肾上腺皮质释放激素(corticotropin releasing hormone,CRH)、促肾上腺皮质激素释放因子(corticotropin releasing factor,CRF)和肾上腺糖皮质激素受体(glucocorticoid reccptor,GR)的影响,探讨热敏灸治疗IBS的可能作用机制。方法:50只SD雄性大鼠按随机数字表随机分为空白组(6只)、模型组(6只)、艾灸组(32只),米非司酮(Mifepristone,RU-486)组(6只),空白组正常饲养,模型组、RU-486组与艾灸组采用慢性不可预见性刺激法建立IBS模型,造模21 d。造模结束后,艾灸组灸命门穴,每日1次,每次40 min,共14天,每隔10 min检测大鼠尾温并记录,根据大鼠尾温变化分热敏灸组和非热敏灸组,两组各随机抽取6只;RU-486组RU-486灌胃处理,空白组与艾灸组灌服同等换算体积生理盐水14 d。观察各组大鼠行为学变化。治疗结束后取下丘脑、肾上腺,-80℃保存待检。结果:(1)与空白组相比,模型组、热敏灸组、非热敏灸组、RU-486组CRF、CRH表达差异均明显升高(P〈0.05,P〈0.01);与模型组相比,热敏灸组、非热敏灸组表达均明显偏低(P〈0.05,P〈0.01),RU-486组表达升高(P〈0.05,P〈0.01);与热敏灸组相比,非热敏灸组、RU-486组表达明显均升高(P〈0.05,P〈0.01)。(2)与空白组相比,模型组、非热敏灸组、RU-486组GR表达均下降,差异均有统计学意义(均P〈0.01),热敏灸组表达无明显差异;与模型组相比,热敏灸组表达偏高,差异有统计学意义(P〈0.05),RU-486组表达偏低(P〈0.01),非热敏灸组无明显差异;与热敏灸组相比,非热敏灸组、RU-486组表达均偏低(P〈0.05,P〈0.01)。结论:热敏灸可能通过改善GR表达,恢复对HPA轴的负反馈作用,从而达到治疗IBS的目的。 Objective: To observe the effect of heat-sensitive moxibustion on CRH and CRF in hypothalamus and GR in adrenal gland of IBS rats and explore the possible mechanism of heat-sensitive moxibustion in treating IBS. Methods: According to random number table,50 SD male rats were randomly divided into the blank group(n = 6),the model group(n = 6),the moxibustion group(n = 30) and the RU-486 group(n = 6). The blank group was fet with normal feeding. The model group,the RU-486 group and the moxibustion group used chronic and non-predictable stimulation method to establish IBS model. After modeling for21 days,the moxibustion group was given moxibustion at Mingmen(DU4),once a day and 40 min,a total of 14 days. The temperature of the tail was recorded every 10 min. According to the change of the temperature of the tail,it could be classifed as the heat-sensitive moxibustion group and the non-heat-sensitive moxibustion group,two groups were randonty selected 6 rats. The RU-486 group was treated with RU-486 by gavage. The blank group and the moxibustion group were given the same volume of normal saline for 14 days. The behavioral changes of rats in each group were observed. After treatment,the hypothalamus and adrenal gland were taken out and stored in the enviorment of-80 ℃. Results:(1) Compared with the blank group,the expressions of CRF and CRH in model group,the heat-sensitive moxibustion group,the non-heat-sensitive moxibustion group and the RU-486 group were significantly different(P〈0. 05,P〈0. 01). Compared with the model group,the expressions in the heat-sensitive moxibustion group and the non-heat-sensitive moxibustion group were significantly lower(P〈0. 05,P〈0. 01) and the expression of the RU-486 group was increased(P〈0. 05,P〈0. 01). Compared with the heat-sensitive moxibustion group,the expressions of the non-heat-sensitive moxibustion group and the RU-486 group were significantly higher(P〈0. 05,P〈0. 01).(2) Compared with the blank group,the expression of GR was decreased in the model group,the non-heat-sensitive moxibustion group and the RU-486 group and the differences were statistically significant(P〈0. 01). There was no significant difference in the heat-sensitive moxibustion group. Compared with the model group,the expression of the heat-sensitive moxibustion group was higher and the difference was statistically significant. The expression of the RU-486 group was lower(P〈0. 01) and there was no significant difference in the non-heat-sensitive group. Compared with the heat-sensitive moxibustion group,the expressions of the non-heat-sensitive moxibustion group and RU-486 group were lower(P〈0. 05,P〈0. 01). Conclusion: Heat-sensitive moxibustion can improve the expressions of GR and restore the negative feedback effect of HPA axis,so as to achieve the goal of IBS treatment.
