胃复方对裸鼠胃癌BGC-823移植瘤及PPARγ表达的影响

Effect of Weifufang on expression of PPARγ in gastric carcinoma BGC-823 transplanted in nude mice

[目的]通过观察胃复方对裸鼠胃癌BGC-823移植瘤及对过氧化物酶增殖激活受体γ(PPARγ)表达的影响,探讨胃复方抑制裸鼠胃癌BGC-823移植瘤的可能机制。[方法]用不同浓度的胃复方药液处理BGC-823移植瘤裸鼠,观察不同剂量胃复方干预后瘤体大小的变化,苏木精-伊红染色观察瘤体组织病理学改变、免疫组织化学和蛋白印迹法检测瘤体组织PPARγ的表达。[结果]胃复方能抑制裸鼠胃癌BGC-823移植瘤的生长,作用于移植瘤裸鼠2周后,5-Fu阳性对照组(5-Fu组)、胃复方低剂量组(低剂量组)、中剂量组、高剂量组的抑瘤率分别为30.67%、19.00%、49.52%、29.36%,以中剂量组最佳;通过免疫组织化学检测,蒸馏水组、5-Fu组、低剂量组、中剂量组、高剂量组PPAR-γ基因表达的IOD值分别为:0.04±0.02、0.05±0.01、0.05±0.01、0.09±0.02、0.06±0.01。胃复方中剂量组与其他各组比较,均差异有统计学意义(均P<0.05)。蛋白印迹法检测PPAR-γ基因表达,蒸馏水组、5-Fu组、低剂量组、中剂量组、高剂量组分别为:0.316 0±0.013 6、0.724 0±0.018 1、0.564 0±0.019 9、0.718 0±0.028 9、0.570 0±0.020 0,蒸馏水组与其他各组比较差异均有统计学意义(均P<0.05);5-Fu组与中剂量组比较,差异无统计学意义(P>0.05);5-Fu组与低剂量、高剂量组比较差异均有统计学意义(均P<0.05)。中剂量组与低、高剂量组比较差异均有统计学意义(均P<0.05)。[结论]胃复方可能通过上调PPARγ表达的途径来抑制胃癌BGC-823移植瘤的生长,且以1倍剂量效果最佳。

[Objective]To explore the possible mechanism of Weifufang in inhibiting BGC-823 transplanted tumor by observing the effect of Weifufang on gastric cancer BGC-823 xenografts in nude mice and the activity of peroxisome proliferator-activated receptorγ（PPARγ）gene.[Methods]The mices of gastric cancer BGC-823 xenografts were treated with different concentrations of Weifufang.The tumor size of thetransplanted tumor was observed with different doses of Weifufang.The histopathological changes of the tumor were observed by HE staining.The expression of PPARγgene was detected by immunohistochemistry and Western blot Methods.[Results]Weifufang could inhibit the growth of BGC-823 xenografts in nude mice by two weeks.The inhibition rate of 5-Fu positive control group,low-dose Weifufang group,middledose Weifufang group and high-dose Weifufang group were：30.67%、19%、49.52%、29.36%.The IOD value of PPAR-γwas detected by immunohistochemistry in distilled water group,the 5-Fu positive control group,low-dose Weifufang group,middle-dose Weifufang group and high-dose Weifufang group were0.04±0.02,0.05±0.01,0.05±0.01,0.09±0.02,0.06±0.01.There was significant difference between each group with middle-dose group（P〈0.05）.The expression of PPAR-γwas detected by Western blotting in distilled water group,the 5-Fu positive control group,low-dose group,middle-dose group and highdose group were 0.316 0±0.013 6,0.724 0±0.018 1,0.564 0±0.019 9,0.718 0±0.028 9,0.570 0±0.020 0.There was significant difference between distilled water group and other groups（P〈0.05）.There was no significant difference between the 5-Fu group and the middle-dose group（P〉0.05）.There were significant difference between 5-Fu group and the low-dose group and high-dose group.There was significant difference between the middle dose group and low and high dose group（P〈0.05）.[Conclusion]Weifufang could inhibit the growth of gastric cancer BGC-823 tumor by up-regulating the expression of PPARγgene,and the effect was best in middle dose group.
