山羊Claudin-1基因编码蛋白的生物信息学分析 被引量:3

摘要 为研究山羊紧密连接相关蛋白Claudin-1基因编码蛋白的组成结构及生物学特性,本试验综合运用多种生物信息学软件对山羊Claudin-1蛋白的理化性质、亲/疏水性、信号肽、跨膜结构域、磷酸化位点、亚细胞定位、蛋白质二级结构及三级结构进行生物信息学分析,并推测与其他物种的生物进化关系。结果表明:山羊Claudin-1基因全长636 bp,共编码211个氨基酸,蛋白质理论大小值为22.865 ku,理论等电点为8.40,呈碱性;不稳定指数为43.82,其编码蛋白属于不稳定的疏水性蛋白;二级结构以α-螺旋为主要结构,比例为54.50%,主要存在于内质网,其次为质膜,在细胞外(包括细胞壁)和空泡中也有少量分布。Claudin-1蛋白具有15个潜在的磷酸化位点,存在信号肽和4个跨膜区。通过进化树分析发现,山羊Claudin-1基因与绵羊和牛的亲缘关系最为密切,这与动物学分类结果相一致,提示Claudin-1基因与胃肠道黏膜上皮选择性屏障存在联系。通过不同的在线预测软件对山羊Claudin-1基因编码蛋白的组成结构及生物学特性进行分析,为深入探讨Claudin-1基因编码蛋白提供理论指导。 This study was aimed to explore the compositional structure and biological characteristics of the claudin-1 gene that encodes proteins related to tight junctions in goat. The physicochemical properties,hydrophilicity/hydrophobicity,signal peptide,transmembrane structure,phosphorylation site,subcellular localization,second structure and tertiary structure of the gene were analyzed by various types of bioinformatics software,and its evolution relationships with other species were speculated. The results indicated that the goat claudin-1 gene was636 bp in full length. It encoded a protein of 211 amino acids. The protein weighed 22. 865 ku. The theoretical p I was 8. 40. The instability index was 43. 82. And the encoded protein was a hydrophobicity protein which was unstable. The secondary structure was mainly Alpha-helix( 54. 50%). The protein was mainly located in the endoplasmic reticulum,and also distributed in plasma membrane,extracellular( including cell wall) and vacuolar. Moreover,there was one signal peptide,four transmembrane domains and 15 phosphorylation sites. In conclusion,the claudin-1 gene in goat had the closest genetic relationship with ovis aries and bos Taurus,based on the analysis of molecular evlution. This was consistent with the results of animal taxonomy,and it suggested that the protein-encoding claudin-1 gene was associated with the gastrointestinal mucosal epithelium selective barrier. The present study provides a theoretical guidance for further research on the claudin-1 that encodes proteins.
