弹性蛋白肽对髓样树突状细胞表面共刺激分子表达及炎症因子分泌的影响 被引量:1

摘要 目的:探讨弹性蛋白肽(EP)对髓样树突状细胞(mDCs)表面共刺激分子表达及炎症因子分泌的影响。方法:诱导小鼠骨髓来源的单个核细胞增殖分化为未成熟mDCs,随机分为空白对照组和EP组,空白对照组不加任何刺激,EP组加入终浓度为1mg/L EP。分别采用流式细胞术和qPCR技术检测mDCs表面共刺激分子CD80和CD86的表达,酶联免疫吸附试验(ELISA)法检测细胞上清液中白介素(IL)-6和转化生长因子(TGF)-β1的浓度。结果:流式细胞仪检测EP组CD80和CD86阳性表达率分别为(53.05±2.49)%、(50.05±3.90)%,显著高于空白对照组的(22.38±5.21)%、(19.33±5.80)%(P<0.05)。EP组CD80和CD86mRNA相对表达量及细胞上清液中IL-6和TGF-β水平均较空白对照组明显升高(P<0.05)。结论:EP可促进小鼠mDCs的活化及相关促炎因子的释放。 Objective:To observe the differences in expression of cell surface co-stimulatory molecules and secretion of inflammatory factors by bone marrow-derived dendritic cells(DCs)in response to elastin peptide(EP).Methods:We cultured bone marrow-derived DCs from BALB/c mice and infected with EP.The activation of myeloid DCs(mDCs)was characterized by studying expression of cell surface co-stimulatory molecules(CD80 and CD86),release of transforming growth factor(TGF)-β1 and interleukin(IL)-12.Results:Flow cytometry analysis and qPCR detection showed significant up-regulation of CD80 and CD86 after treatment with EP(P〈0.05).Meanwhile,ELISA revealed that the IL-6 and TGF-β1 levels in cell supernatant was conspicuous elevated(P〈0.05).Conclusion:EP could promote the activation of mDCs in mice and the release of related pro-inflammatory factors.
