变形链球菌提取物对人牙髓细胞TOLL样受体2表达的影响

Effect of extracts from Streptococcus mutans on the expression of Toll-like receptor 2 in human dental pulp cells

目的:探讨变形链球菌提取物对人牙髓细胞(hDPCs)TOLL样受体2(TLR2)表达的影响。方法:超声破碎法提取变形链球菌的提取物,应用体外培养技术培养hDPCs,将变形链球菌提取物按10mg/L和100mg/L浓度分组,以等体积的培养基作为空白对照组,分别作用于hDPCs 0h、24h、48h。免疫组化法检测hDPCs TLR2的表达和分布情况,实时荧光定量PCR(qPCR)检测TLR2mRNA的表达水平。结果:免疫组化结果显示,10mg/L和100mg/L组各时间点均有TLR2蛋白表达,均明显高于空白对照组(P<0.05),10mg/L组与100mg/L组各作用时间点比较,差异无统计学意义(P>0.05)。qPCR结果显示,10mg/L和100mg/L组刺激24h、48h后,TLR2 mRNA相对表达量明显升高,并且随刺激时间的延长表达量逐渐增加,均高于空白对照组(P<0.05)。结论:变形链球菌提取物可促使hDPCs TLR2表达上调,提示TLR2可能参与了牙髓的天然免疫反应。

Objective：To investigate the difference of Toll-like receptor 2（TLR2）expression in human dental pulp cells（hDPCs）in response to extracts fromStreptococcus mutans（ESM）.Methods：We prepare the ESM by sonication and centrifugation.hDPCs were cultured in vitro and respectively infected with 10 mg/L and 100 mg/L of ESM for 24 hand 48 h.The mRNA and protein expressions of TLR2 were determined by immunohistochemistry and qPCR,respectively.Results：The expression of TLR2 mRNA and protein was markedly up-regulated in hDPCs at 24 hand 48 hafter 10 mg/L and 100 mg/L of ESM infection.No significant difference was noted in the TLR2 expression between low and high dose ESM groups.The TLR2 mRNA level was increased with the time prolonged（P〈0.05）.Conclusion：ESM could enhance the expression of TLR2 in hDPCs,indicating that the TLR2 might be involved in the innate immune response of dental pulp cells.
