摘要
目的:探讨变形链球菌提取物对人牙髓细胞(hDPCs)TOLL样受体2(TLR2)表达的影响。方法:超声破碎法提取变形链球菌的提取物,应用体外培养技术培养hDPCs,将变形链球菌提取物按10mg/L和100mg/L浓度分组,以等体积的培养基作为空白对照组,分别作用于hDPCs 0h、24h、48h。免疫组化法检测hDPCs TLR2的表达和分布情况,实时荧光定量PCR(qPCR)检测TLR2mRNA的表达水平。结果:免疫组化结果显示,10mg/L和100mg/L组各时间点均有TLR2蛋白表达,均明显高于空白对照组(P<0.05),10mg/L组与100mg/L组各作用时间点比较,差异无统计学意义(P>0.05)。qPCR结果显示,10mg/L和100mg/L组刺激24h、48h后,TLR2 mRNA相对表达量明显升高,并且随刺激时间的延长表达量逐渐增加,均高于空白对照组(P<0.05)。结论:变形链球菌提取物可促使hDPCs TLR2表达上调,提示TLR2可能参与了牙髓的天然免疫反应。
Objective:To investigate the difference of Toll-like receptor 2(TLR2)expression in human dental pulp cells(hDPCs)in response to extracts fromStreptococcus mutans(ESM).Methods:We prepare the ESM by sonication and centrifugation.hDPCs were cultured in vitro and respectively infected with 10 mg/L and 100 mg/L of ESM for 24 hand 48 h.The mRNA and protein expressions of TLR2 were determined by immunohistochemistry and qPCR,respectively.Results:The expression of TLR2 mRNA and protein was markedly up-regulated in hDPCs at 24 hand 48 hafter 10 mg/L and 100 mg/L of ESM infection.No significant difference was noted in the TLR2 expression between low and high dose ESM groups.The TLR2 mRNA level was increased with the time prolonged(P〈0.05).Conclusion:ESM could enhance the expression of TLR2 in hDPCs,indicating that the TLR2 might be involved in the innate immune response of dental pulp cells.
