摘要
目的研究小檗碱(BBR)对去卵巢(OVX)小鼠学习记忆能力的影响并探讨可能机制。方法30只雌性C57BL/6小鼠随机分成假手术组(SHAM组),去卵巢组(OVX组),去卵巢+小檗碱组(OVX+小檗碱组)3组,治疗8w,通过Morris水迷宫测试认知能力,采用Wester blot检测小鼠大脑组织中Bcl-2和Bax蛋白的表达。结果与OVX组相比,BBR治疗后OVX小鼠的逃避潜伏期明显缩短(P<0.05),穿越平台次数明显增多(P<0.05);与SHAM组比较,OVX组小鼠Bcl-2表达降低和Bax表达增加(P<0.05),BBR治疗后,OVX小鼠Bcl-2表达增加和Bax表达降低(P<0.05)。结论 BBR能改善OVX小鼠的学习记忆能力,其机制可能与大脑组织中Bcl-2增加和Bax降低有关。
Objective To observe the effect of berberine(BBR) on ovariectomized mice in cognitive function and explore possible mechanisms. Method 30 female C57 BL/6 mice were randomly divided into three groups: SHAM, OVX, OVX+BBR. After 8 weeks, cognitive ability was detected by Morris water maze test, and Western blot was used to detect the expression of Bcl-2 and Bax in the brain tissue. Results Compared with OVX group, the avoidance latency period of OVX mice treated with BBR was significantly decreased(P〈0.05), and the number of cross-platform times was significantly increased(P〈0.05). Comparison with SHAM group, the expression of Bcl-2 decreased and the expression of Bax increased in OVX group(P〈0.05). After BBR treatment, the expression of Bcl-2 increased and the expression of Bax decreased in OVX mice(P〈0.05). Conclusion BBR can improve the cognitive ability of OVX mice, and its mechanism may be related to the increase of Bcl-2 and the decrease of Bax in brain tissue.
