摘要
探讨了一株植物乳杆菌在高密度培养过程中的适宜限制性底物的浓度、比例和补料流加模式。研究表明:发酵液初始葡萄糖浓度为15 g/L,初始蛋白胨浓度为14 g/L。补料液的限制性底物为葡萄糖和蛋白胨,它们的浓缩液体积比例为5∶7;在补料的同时添加碱液和乙酸钠中和过多的酸。同时探讨了不同的补料方式对活菌数的影响,确定葡萄糖反馈流加能够维持一定的低糖浓度,获得较高的活菌数,达到3.1×10~9CFU/m L,较之原来提高了4倍。
The high-density culture of Lactobacillus plantarum by batch feeding was researched. The results showed that: the initial concentration of glucose and peptone in fermentation broth was 15 g/L and 14 g/L,respectively; the restrictive substrates of feeding liquid were glucose and peptone,and the volume ratio of their concentrated solution was 5 ∶ 7; in the course of batch feeding,sodium hydroxide and sodium acetate were added to neutralize the excessive acids in the solution; the glucose feedback feeding mode could maintain a low glucose concentration,and obtain higher viable count( 3.1× 109 CFU/m L),which was 5 times that in the normal culture.
