金刚烷胺抑制乙脑病毒感染引起小胶质细胞炎症反应的作用 被引量:3

The effect of amantadine on inhibition of inflammatory reactions of microglia induced by Japanese encephalitis virus infection

摘要 目前金刚烷胺被(areantadine,Ama)用作一种抗病毒和抗帕金森药物,可抑制小胶质细胞的炎症激活。为验证金刚烷胺是否会抑制乙脑病毒(Japanese encephalitis virus,JEV)感染小胶质细胞所引起的炎症反应.本试验利用Ama处理小鼠小胶质瘤BV-2细胞系后使用JEV感染细胞,测定细胞中炎性因子的表达,并与单独使用JEV处理BV-2细胞组进行对比分析。结果表明,Ama可以显著降低JEV感染引起的5种炎性因子的表达,MCP-1(62.17%)、IL-6(62.07%)、TNF-α(59.67%)、IL-18(63.16%)以及IL-10(34.24%)。本试验为流行性乙型脑炎的临床治疗提供了理论依据。 Amantadine is currently used as an antiviral drug and an antiparkinsonian drug,and it can also inhibit the inflammatory activation of microgtia. In order to verify whether amantadine will inhibit the inflammatory response of microglia infected by Japanese encephalitis virus. In this re- search,BV-2 cells pretreated with or without amantadine, followed by infection with Japanese en- cephalitis virus,and then relative inflammatory cytokines were determined. We found that arnanta- dine significantly reduced the expression of five inflammatory cytokines induced by Japanese en- cephalitis virus infection: MCP-1 (62.17%),IL-6(62.07%),TNF-α (59. 67%),IL-1β(63.16%)and IL-10(34.24%). MCP-1, IL-6, TNF-α and IL-1β are all proinflammatory cytokines and their de- creases indicate that amantadine effectively inhibits inflammatory responses. Especially, the de- crease of IL-10 indicates that the degree of inflammatory responses has been effectively alleviated.This research provides a theoretical basis for the clinical treatment of Japanese encephalitis.
