洗心汤对Aβ1-42诱导的海马神经元线粒体功能的影响

Effects of Xixin Decoction on Mitochondrial Function of Hippocampal Neurons Induced by Aβ1-42

目的：观察名方洗心汤对Aβ（1-42）损伤的海马神经元线粒体的调控作用,探讨名方洗心汤防治阿尔茨海默病（Alzheimer＇s disease,AD）的可能作用机制。方法：提取Wistar新生鼠海马神经元并培养,分正常对照组,Aβ模型组,低、中、高浓度治疗组;Annexin V/PI法检测细胞凋亡率;JC-1荧光法检测线粒体膜电位;流式细胞仪技术和免疫荧光技术分别检测ROS与Cyt-c的蛋白基因表达。结果：洗心汤含药脑脊液对Aβ（1-42）诱导所致细胞凋亡具有显著保护作用。结论：名方洗心汤能够修复线粒体功能,从而提高Aβ（1-42）诱导细胞的存活率。

Objective： To observe the effect of Xixin Decoction（ XXD） on mitochondrial fuction in hippocampal neurons injured by Aβ（1-42） and explore the possible mechanism. Methods： We primarily cultured hippocampal neurons of Wistar rats. The neurons were randomly divided into normal group,Aβ（1-42） model group,and low,moderate,high dose XXD groups. Apoptosis rate was detected by Annexin V/PI double staining flow cytometry. JC-1 fluorescence was used to detect mitochondrial membrane potential.Flow cytometry detected ROS. Immunofluorescence technique was used to detect the expression of Cyt-c protein gene. Results：The cerebrospinal fluid with XXD has a significant protective effect on Aβ（1-42）-induced apoptosis. Conclusion： XXD can repair mitochondrial function and the survival rate of Aβ（1-42） induced cells has been increased.
