补脾强力复方对实验性自身免疫性重症肌无力大鼠下丘脑CRHmRNA及蛋白表达的影响 被引量:2

摘要 目的:观察补脾强力复方对实验性自身免疫性重症肌无力(EAMG)大鼠下丘脑CRHmRNA及蛋白表达的变化,探讨补脾强力复方治疗EAMG的作用机制。方法:将Lewis大鼠作为正常对照组,采用免疫诱导注射鼠源性AchR-α亚基97-116肽段免疫Lewis大鼠复制EAMG模型,将造模成功的EAMG大鼠随机分为模型组、强的松组(5.4 mg/kg)、补脾强力复方大、中、小剂量组(分别为9.49 g/kg,7.12 g/kg,4.75 g/kg),每日灌胃治疗1次,连续给药4周后分别采用RT-Q-PCR技术、免疫组化法观察各组大鼠下丘脑CRHmRNA及蛋白表达变化。结果:EAMG模型组大鼠下丘脑CRHmRNA及蛋白表达较正常组均明显降低(P〈0.01);补脾强力复方大、中、小剂量组下丘脑CRHmRNA及蛋白较EAMG模型组及强的松组相对表达量增加(P〈0.05)。结论:补脾强力复方能上调EAMG大鼠下丘脑CRHmRNA及蛋白表达,这可能是补脾强力复方治疗EAMG大鼠机制之一。 Objective: To research the effect of Compound Bupi Qiangli Decoction on the mRNA and protein expression levels of hypothalamus CRH in experimental autoimmunemyasthenia gravis( EAMG) rats and investigate mechanism of Compound Bupi Qiangli Decoction in EAMG rats. Methods: Lewis rats were as the control group,immunized with a synthetic peptide corresponding to region 97-116 of the rat acetylcholine receptor-α subunit( R97-116) to establish EAMG rat models. The successful models were randomly divided into 5 groups: EAMG model group,prednisone group( 5. 4 mg/kg),Compound Bupi Qiangli Decoction low,medium,high dose groups( dosage 9. 49 g/kg,7. 12 g/kg,4. 75 g/kg). The rats were intragastrically administrated once a day and continously for 4 weeks. And then the mRNA and protein expression levels of hypothalamus CRH were observed by RT-Q-PCR and Immunohistochemistry. Results: Compared with the Lewis control group before treatment,the mRNA and protein expression levels of hypothalamus CRH in model rats decreased more,showing significance between groups( P〈0. 01). The results of the groups with low,medium and high dose of Compound Bupi Qiangli Decoction increased more compared with those of both Prednisone group and EAMG model group and the results showed significance( P〈0. 05) Conclusion: Compound Bupi Qiangli Decoction can increase the mRNA and protein expression of CRH in hypothalamus effectively,which may be one of the important therapeutic mechanisms of Compound Bupi Qiangli Decoction in treatment of myasthenia gravis.
