光老化人真皮微血管内皮细胞模型的建立及桃红四物汤的保护作用 被引量:2

Establishment of Photoaging Human Dermal Microvascular Endothelial Cell Model and The Protective Effect of Taohong Siwu Decoction

摘要 目的:建立人真皮微血管内皮细胞光老化模型,并观察桃红四物汤对其影响。方法:体外培养人真皮微血管内皮细胞,将其接种于96孔板中,分为UVA模型组、空白血清组、低剂量含药血清组、中剂量含药血清组、高剂量含药血清组5组,各组细胞接种后,培养24 h,以5 J/cm2的照射剂量对细胞进行照射,于照射后24 h在倒置光学显微镜下观察细胞损伤的形态学变化,采用CCK-8法检测各组细胞增殖活性,评价桃红四物汤含药血清抗HDMEC细胞光老化作用。结果:与空白对照组比较,空白血清组及不同浓度桃红四物汤含药血清组细胞活性显著降低,有统计学差异(P〈0.01);与空白血清组比较,不同浓度的桃红四物汤含药血清组细胞增值活性均显著升高(P〈0.01);与低剂量含药血清组比较,中高剂量含药血清组细胞活性显著升高。结论:桃红四物汤对光老化人真皮微血管内皮细胞具有一定的保护作用。 Objective: To establish the photoaging model of HDMECs and observe the effect of Taohong Siwu Decoction. Methods: The human dermal microvascular endothelial cells cultured in vitro and vaccinated in 96-well plates,were divided into 5 groups: UVA model group,blank serum group,low-dose drug serum group,medium-dose drug serum group,high dose drug serum drug. After each group's cells was vaccinated,they were trained for 24 hours. The cells were irradiated with 5 J/cm2 exposure dose. After 24 hours,we observed the morphological changes of cell damage under the inverted light microscope. The CCK-8 method was used to detect each group's cells proliferation activity and we evaluated the effect of Taohong Siwu Decoction on photoaging HDMECs. Results: Compared with blank control group,the cell proliferation activities of blank serum group and serum containing different concentrations of Taohong Siwu Decoction were significantly lower( P〈0. 01). Compared with blank serum group,the cell proliferation activities of the serum containing different concentrations of Taohong Siwu Decoction were significantly higher( P〈0. 01). Compared with low dose group,medium and high dose groups' cell proliferation activities were significantly higher( P〈0. 01). Conclusion: Taohong Siwu Decoction has a protective effect on the photoaging HDMECs.
