云南壮族、苗族、纳西族人群HNA-3基因多态性分布

Polymorphism distribution of the HNA-3 gene among Zhuang,Miao and Naxi populations in Yunnan province,China

摘要 目的了解云南地区献血者中少数民族人群的人类中性粒细胞抗原-3(HNA-3)的分布频率。方法从2012年1月—2014年1月在昆明血液中心献血的云南本地无血缘关系的健康献血者外周血标本中,筛选出壮族196(人)份、苗族209(人)份和纳西族187(人)份,采用血液基因组DNA提取试剂盒提取其DNA,以PCR-SBT方法对其HNA-3作基因分型,利用SPSS 20.0统计学软件分析3个民族的HNA-3等位基因分布频率。结果本组云南壮族、苗族和纳西族献血人群的HNA-3a/a基因型频率分别为0.59(115/196)、0.48(101/209)和0.45(85/187),HNA-3a/b分别为0.35(69/196)、0.43(90/209)和0.46(86/187),HNA-3b/b分别为0.06(12/196)、0.09(18/209)和0.09(16/187),HNA-3a分别为0.76(298/392)、0.66(276/418)和0.68(254/374),HNA-3b分别为0.24(94/392)、0.34(142/418)和0.32(120/374);云南壮族HNA-3b/b基因型频率明显低于云南苗族和纳西族人群(P〈0.05)。结论本组云南3个目标民族人群中HNA-3等位基因频率的分布存在多态性;云南本地苗族和纳西族人产生抗-HNA-3a的风险可能要高于壮族人。 Objective To investigate the frequency distribution of the human neutrophil antigen-3( HNA-3) among the minority populations in Yunnan blood donors.Methods Target samples were acquired from unrelated healthy Yunnan blood donor samples collected during Jan,2012——Jan,2014,which included 196 Zhuang individuals,209 Miao individuals and 187 Naxi individuals. The DNA of these donors were extracted by blood DNA extraction kits. PCR-SBT was applied for HNA-3 genotyping,and the SPSS 20. 0 software was used for the statistical analysis of the HNA-3 allele frequency. Results The frequencies of the HNA-3 a/a alleles were 0. 59( 115/196),0. 48( 101/209),0. 45( 85/187) in the Zhuang,Miao and Naxi samples,respectively. The frequencies of the HNA-3 a/b alleles were 0. 35( 69/196),0. 43( 90/209) and 0. 46( 86/187),respectively. The frequencies of the HNA-3 b/b alleles were 0. 06( 12/196),0. 09( 18/209) and 0. 09( 16/187),respectively. The frequencies of the HNA-3 a alleles were 0. 76( 298/392),0. 66( 276/418) and 0. 68( 254/374),respectively. The frequencies of the HNA-3 b alleles were 0. 24( 94/392),0. 34( 142/418) and 0. 32( 120/374),respectively. The genotype HNA-3 b/b in the Zhuang samples were significantly lower than that in the Miao and Naxi samples( P 〈0. 05). Conclusion The distribution of the HNA-3 is polymorphic among the three minority populations in Yunnan province.The risk of HNA-3 a alloantibodies in the Miao and Naxi populations may be higher than that of the Zhuang populations.
