摘要
目的:观察少腹逐瘀汤对子宫内膜异位症模型大鼠氧化应激状态的影响,探讨少腹逐瘀汤治疗内异症的机制。方法:58只有正常动情周期的SD雌性大鼠,随机选择10只作为对照组,剩余大鼠采用自体移植法复制内异症大鼠模型,造模成功的38只大鼠随机分为模型组及治疗组,治疗组给予少腹逐瘀汤灌胃,给药4周。测量治疗前后子宫内膜异位病灶体积变化,检测治疗后血浆、卵巢组织及异位病灶组织中,脂质过氧化物标志物丙二醛(MDA)水平及抗氧化酶SOD、GSH-PX活性。结果:治疗后,治疗组异位病灶体积较治疗前明显缩小(P<0.01),且明显小于模型组(P<0.01);模型组大鼠血浆、卵巢组织中MDA水平较对照组明显升高(P<0.01);治疗组大鼠血浆、卵巢组织、异位病灶组织中的SOD、GSH-PX活性较模型组明显升高(P<0.01),MDA水平较模型组明显降低(P<0.01)。结论:内异症模型大鼠存在氧化应激损伤,少腹逐瘀汤可以提高模型大鼠的抗氧化能力,减轻氧化应激反应,缩小异位病灶体积。
Objective: To observe the effect of Shaofu Zhuyu Decoction (SFZY) on oxidative stress makers in endometriosis rats, and to explore the mechanisms of SFZY for treating endometriosis (EMT). Methods: 10rats were randomly assigned to con- trolled group from 58female SD rats with regular estrous cycles, the rest rats underwent surgical induction of endometriosis with autotransplantation. 38rats with successful ectopic implants were divided into two groups: model group and treated group. SFZY was administered to rats in the treated group by gastrogavage. The volume of endometriotic lesions was measured after the first and second laparotomy. In addition, the serum, ovaries and endometrial explants were analyzed for MDA, a maker of lipid peroxida- tion and antixidant enzymes SOD and GSH-PX afer treatment. Results: After treatment, the mean explants volume in the treat- ment group was reduced than before treatment ( P〈0. 01 ), the mean explants volume in the treatment group was significantly low- er than that in model group ( P〈0.01 ). In model group, the level of MDA in serum and ovaries significantly increased when com- pared with the controlled group (P〈0. 01 ). In the treated group, the activities of SOD and GSH-PX in serum, ovaries and ex- plants were significantly higher and the level of MDA was significantly lower than that in model group ( P〈0. 01 ). Conclusions : There is a oxidative stress status in endometriosis rats. SFZY can enhance antioxidant capacity to reduceoxidative stress and de- crease the size of endometriotic lesions in EMT rats.
