促愈熏洗方对人皮肤成纤维细胞的增殖、凋亡和迁移的影响

Effect of Cuyu Xunxi Recipe on Proliferation,Apoptosis and Migration for Human Skin Fibroblasts

目的:研究外用中药"促愈熏洗方"对人皮肤成纤维细胞的增殖、凋亡和迁移的调节作用。方法:制备"促愈熏洗方"水提取物,对通过加工萃取获得的乙酸乙酯、水、正丁醇三个部分进行研究。以HFF-1细胞系为研究对象,将不同浓度的各萃取部分干预HFF-1细胞不同时间后,应用CKK-8法检测细胞的增殖情况,流式细胞术检测HFF-1细胞的凋亡情况,应用细胞划痕实验检测细胞的迁移情况。结果:乙酸乙酯萃取部分能够促进HFF-1的增殖能力和迁移能力,31.25μg/ml作用24小时时作用最强;水萃取部分能够促进HFF-1的增殖能力和迁移能力,且延迟晚期凋亡,31.25μg/ml作用24小时时作用最强;正丁醇萃取部分能够促进HFF-1的增殖能力和迁移能力,抑制细胞凋亡,31.25μg/ml作用24小时时作用最强。结论:"促愈熏洗方"水提取物能够促进HFF-1的增殖能力和迁移能力,抑制细胞凋亡,部分阐释了临床应用"促愈熏洗方"促进痔和肛瘘患者手术创面愈合的作用机制。

Objective ： To observe the effect of Cuyu Xunxi Recipe on proliferation, apoptosis and migration of human skin fi- broblasts. Methods： We researched the three parts： ethyl acetate, water and n-butanol obtained by extracting from Cuyu Xunxi Recipe. HFF-lce11 line was regarded as the research object. With the different concentration of the three parts after different time, we applied CKK-Smethod to detect cell proliferation, flow cytometry to detect cell apoptosis of HFF-1, scratch test to de- tect cell migration. Results： The ethyl acetate promoted the proliferation and the migration of HFF-1. The part of 31.25p^g/mlhad the strongest effect on 24h. The water promoted the proliferation and the migration of HFF-land delayed the late apoptosis. The part of 31.25μg/ml also had the strongest effect on 24h. The n-butanol promoted the proliferation and the migration of HFF -land inhibited apoptosis. At the same time, the part of 31. 25μg/mt had the strongest effect on 24h. Conclusion： We have found the aqueous extract of Cuyu Xunxi Recipe can promote the proliferation and migration of HFF-1, inhibiting cell apoptosis. The part results have illustrated the mechanism of promoting action of hemorrhoids and anal fistula patients surgical wound healing.
