STAT1与CD74/CD44在结肠癌细胞中协同调控上皮-间质转化机制研究 被引量:2

摘要 目的:研究信号转导和转录激活子1(STAT1)与CD74/CD44在结肠癌细胞上皮-间质转化调控中的相互作用关系。方法:构建基因STAT1和CD74/CD44真核表达质粒以及上皮-间质转化酪氨酸激酶受体-2(DDR2)启动子荧光素酶报告基因表达质粒,真核表达质粒经转染后,利用免疫共沉淀技术探讨基因STAT1和CD74/CD44的相互作用,通过荧光素酶分析技术分析基因STAT1和CD74/CD44对上皮-间质转化酪氨酸激酶受体-2(DDR2)启动子是否存在协同激活作用。结果:STAT1可将CD74/CD44沉淀下来,CD74/CD44可将STAT1沉淀下来,上皮-间质转化酪氨酸激酶受体-2(DDR2)启动子荧光表达质粒与两者共转染比单独转染STAT1或CD74/CD44对上皮-间质转化酪氨酸激酶受体-2(DDR2)启动子的激活作用更强。结论:STAT1和CD74/CD44之间具有相互作用,在结肠癌发生及发展过程中起着重要的协同作用。
