复方三草滴丸防治大鼠肾性贫血实验研究

Experimental Study on Compound Three Herbs Dripping Pills in the Treatment of Rats with Renal Anemia

目的观察复方三草滴丸对大鼠肾性贫血的防治作用及作用机制,为复方三草滴丸的剂型改造和临床应用提供依据。方法连续15d给大鼠皮下注射庆大霉素造模,同时给予复方三草滴丸溶解后灌胃治疗,观察各组大鼠血肌酐(SCr)、尿素氮(BUN)、外周血红细胞(RBC)、血红蛋白(Hb)和促红细胞生成素(EPO)水平,并作肾组织病理切片。结果复方三草滴丸能降低肾性贫血模型大鼠血BUN水平(P<0.01),提高外周血RBC及Hb水平(P<0.05),血SCr有下降,外周血EPO有提升,但均无明显差异(P>0.05),减少肾单位坏死,并减轻肾组织炎性浸润。结论复方三草滴丸对庆大霉素所致肾性贫血有一定治疗作用,其作用机制可能与延缓肾坏死、改善肾功能和增加EPO水平有关。

Objective To observe the preventive and therapeutic effects and mechanism of Compound Three Herbs Dripping Pills on renal anemia of rats, in order to provide a theoretical basis for its clinical application. Methods For establishing the models, the rats were injected with gentamicin subcutaneously for 15 d. At the same time, Compound Three Herbs Dripping Pills were given intragastrically. The levels of serum creatinine（ SCr）,urea nitrogen（ BUN）,the peripheral blood red blood cells（ RBC）,hemoglobin（ Hb） and erythropoietin（ EPO） of the rats were observed,and the pathological section of nephridial tissue was made. Results Compound Three Herbs Dripping Pills could reduce the BUN level of the rats with renal anemia（ P〈0. 01）, and increase the levels of RBC and Hb in peripheral blood（ P〈0. 05）. This drug could reduce the SCr,increase the EOP in peripheral blood,but the difference was not statistically significant（ P〉0. 05）. The drug also could reduce the nephron necrosis and the inflammatory infiltration of renal tissue.Conclusion Compound Three Herbs Dripping Pills have certain therapeutic effects on gentamicin-induced renal anemia. The mechanism may be related to delaying renal necrosis,improving renal function and increasing the level of EPO.
