超声浸提-高效液相色谱法测定刺三加根和茎中没食子酸含量 被引量:6

摘要 建立超声浸提-高效液相色谱法快速测定不同地区刺三加根和茎中没食子酸含量的方法。选择ZORBAX SB-C18(4.6 mm×250 mm,5μm)色谱柱,以甲醇-水-冰醋酸(44∶55.6∶0.4,v/v/v)溶液为流动相,检测波长为272 nm,流速1.0 m L/min,柱温25℃。没食子酸在0.10~5.18μg/m L范围内与峰面积呈良好的线性关系,线性回归方程为A=27.5078C–2.6833(C:μg/m L),r=0.9990;刺三加根和茎中没食子酸平均加标回收率分别为99.96%(RSD=0.86%,n=6)和99.83%(RSD=0.60%,n=6)。该方法操作简便快捷、灵敏,结果准确,且具有良好的稳定性、重复性和回收率,可为刺三加中没食子酸的质量控制和评价提供科学依据。 A new method for the rapid determination of gallic acid content in Acanthopanan trifoliatus roots and stems from different habitats was established by ultrasonic extraction and high performance liquid chromatography(HPLC). Agilent ZORBAX SB-C18(4.6 mm×250 mm,5 μm)was used,methanol and 0.4% acetic acid was used as mobile phase,flow rate was 1.0 m L/min,the detection wavelength was 272 nm and the column temperature was 25 ℃. There was a good linear relationship between the concentration of gallic acid and peak area within 0.10-5.18 μg/m L. The linear regression equation of the calibration curve was A = 27.5078 C – 2.6833(C :μg/m L)with a correlation coefficient of 0.9990. The average recovery rate of galic acid in Acanthopanan trifoliatus roots and stems was 99.96%(RSD=0.86%,n=6)and 99.83%(RSD=0.60%,n=6). As a result,the analysis method,which has good stability,repeatability,and recovery,is simple,rapid,accurate and sensitive,thus,it can provide scientific basis for the quality control and evaluation of gallic acid in Acanthopanan trifoliatus.
