摘要
以水解液中木二糖、木三糖比例为考察指标,筛选适合于工业生产制备高纯度低聚木糖的产木聚糖酶菌株。优化低聚木糖制备工艺条件,经单因素试验及Box-Benhnken响应面分析,确定最佳水解条件。结果表明:当底物质量分数2.4%,加酶量5 U/m L,水解时间6.5 h,水解温度57℃时,木二糖、木三糖比例达到86.23%,较优化前提高了37.46%。
The xylanase producing strain of high purity xylo-oligosaccharide was screened by the ratio of two xylose and three sugar in the hydrolysate. The optimum preparation conditions of xylo-oligosaccharides were optimized, and the best hydrolysis conditions were determined by single factor test and Box-Benhnken response surface analysis. The results showed that when the concentration of substrate was 2.4%, the amount of enzyme was 5 U/m L, the time of hydrolysis was 6.5 h, and the hydrolysis temperature was 57, the proportion of wood two sugars and wood three sugar reached86.23%, which was improved by 37.46% compared with that before the optimization.
