miRNA-27b在子痫前期胎盘中的表达及丹参对其表达的影响 被引量:4

摘要 目的:探讨微小RNA-27b(miRNA-27b)在子痫前期中的表达及丹参对其表达的影响。方法:胎盘组织标本取自于20例重度子痫前期和20例轻度子痫前期病例。选择20名健康产妇的胎盘组织作为对照组。采用实时荧光定量技术(q RTPCR)检测miRNA-27b、瘦素(leptin)m RNA的表达水平,采用瞬时转染法对miRNA-27b进行处理,并采用q RT-PCR、流式细胞术分别检测leptin m RNA水平及人滋养细胞(HTR-8)细胞凋亡情况。结果:重度子痫前期组胎盘组织中miRNA-27b、leptin m RNA的表达水平高于轻度子痫前期组和对照组,且轻度子痫前期组miRNA-27b、leptin m RNA的表达水平高于对照组(P<0.05)。HTR-8细胞中miRNA-27b mimics组中miRNA-27b、leptin m RNA的表达明显较Con组和NC组上调(P<0.05),miRNA-27b inhibitor组中miRNA-27b的表达较Con组和NC组下调(P<0.05);Con组和NC组中miRNA-27b、leptin m RNA的表达水平差异均无统计学意义(P>0.05);miRNA-27b mimics组中HTR-8细胞凋亡水平高于Con组和NC组(P<0.05),miRNA-27b inhibitor组中HTR-8细胞凋亡水平低于Con组和NC组(P<0.05)。重度子痫前期组和轻度子痫前期组胎盘组织中miRNA-27b、leptin m RNA的表达水平均较丹参作用前下降(P<0.05)。结论:miRNA-27b、leptin m RNA在子痫前期胎盘组织中表达上调,miRNA-27b可促进滋养细胞的凋亡,miRNA-27b对leptin起正性调控作用,两者在子痫前期的发生及进展中可能共同起作用。丹参可抑制子痫前期胎盘组织中miRNA-27b的表达,并下调leptin的表达。 Objective: To investigate the expression of miRNA-27 b in placenta of preeclampsia patients,and the effects of Salvia miltiorrhiza on its expression. Methods: Twenty placental tissue samples from severe preeclampsia patients,20 placental tissue samples from mild preeclampsia and 20 placental tissue samples from healthy placental tissue were divided into the severe preeclampsia group,mild preeclampsia group and control group,respectively. The expression levels of miRNA-27 b and leptin m RNA were detected using the real-time fluorescence quantitative PCR( q RT-PCR). The miRNA-27 b was processed using transient transfection,and the leptin m RNA level and human sertoli cells HTR-8 apoptosis were detected using the q RT-PCR and flow cytometry,respectively. Results: The expression levels of miRNA-27 b and leptin m RNA in severe preeclampsia group were higher than those in mild preeclampsia group and control group,and the expression levels of miRNA-27 b and leptin m RNA in mild preeclampsia were higher than those in control group( P〈0. 05). The expression levels of miRNA-27 b and leptin m RNA in miRNA-27 b mimics group significantly increased compared with the Con group and NC group( P〈0. 05),and the expression level of miRNA-27 b in miRNA-27 b inhibitor group decreased compared with the Con group and NC group( P〈0. 05). The differences of the expression levels of miRNA-27 b and leptin m RNA between Con group and NC group were not statistically significant( P〉0. 05). The apoptosis levels of HTR-8 cells in miRNA-27 b mimics group and miRNA-27 b inhibitor group were higher and lower than that in Con group and NC group,respectively( P〈0. 05). The expression levels of miRNA-27 b and leptin m RNA in severe preeclampsia group and mild preeclampsia group decreased compared with before Salvia miltiorrhiza treatment( P〈0. 05). Conclusions: The expression levels of miRNA-27 b and leptin m RNA in preeclampsia placenta increase,miRNA-27 b can promote the apoptosis of sertoli cells. miRNA-27 b plays a positive regulation to leptin,both may work together in the development of preeclampsia. Salvia miltiorrhiza can inhibit the expression of miRNA-27 b and leptin in preeclampsia.
