摘要
本研究以采自黄山地区海拔350~1 850 m的马尾松群体、黄山松群体及其渐渗群体的416个个体为材料,利用筛选的10对SSR引物研究了群体遗传多样性、遗传结构及群体间的渐渗。结果表明:10对SSR引物检测到的等位基因共37个,各位点等位基因数(A)的范围为2~7个,Shannon指数(I)平均为1.0301;按16组海拔高度来分析,10对SSR引物扩增的平均等位基因数为3.1个,平均有效等位基因数(Ne)约为2.4个,Shannon指数(I)平均为0.849 5。根据16组海拔高度内松树群体的等位基因频率变化及16组海拔区间群体的遗传距离和海拔差进行Mantel检测分析,发现黄山松基因向渐渗群体渐渗的强度高于马尾松。以上结果显示黄山松群体与采集的416个个体中大部分个体亲缘关系较近,这说明黄山松在不同海拔松树群体的基因渐渗中起主导作用。
416 natural pinetree samples of Pinus massoniana,Pinus taiwanensis and introgressive populations from Huangshan area(altitude from 350 m to 1 850 m) were selected as the material,and ten pairs of screened SSR primers were used to study the genetic diversity,genetic structure and the introgressive hybridization between populations.The results showed that a total of 37 alleles were determined by 10 pairs of SSR primers,and the alleles of each locus(A) were 2~7.The mean Shannon's information index(Ⅰ) was 1.030 1.According to the analysis of 16 sets of altitude,the average number of alleles amplified by 10 pairs of SSR primers was 3.1,the average effective allele number(Ne) was about 2.4,and the average Shannon index(Ⅰ) was 0.849 5.Based on the variation of allelic frequency of pine populations in 16 groups of altitude,and the genetic distance and altitude difference among 16 groups of altitude,the Mantel detection was conducted.The results indicated that the gene introgression of P.taiwanensis was stronger than P.massoniana.The above findings showed that P.taiwanensis had close relationship with most of the individuals of the 416 individuals collected,suggesting that P.taiwanensis might play a leading role in gene introgression of pine populations at different altitudes.
