不同品种软枣猕猴桃愈伤组织诱导及植株再生 被引量:10

Callus Induction and Plantlet Regeneration of Actinidia arguta with Different Varieties

摘要 以软枣猕猴桃(Actinidia arguta)品种"魁绿"和"佳绿"的无芽茎段、叶片、叶柄为外植体,研究外植体及生长调节剂组合对软枣猕猴桃愈伤组织诱导及植株再生的影响,从而建立"魁绿"和"佳绿"的离体再生体系。结果表明:6-BA对软枣猕猴桃愈伤组织的诱导效果好于KT,"魁绿"和"佳绿"叶片分别在MS+2,4-D0.5 mg/L+6-BA 2 mg/L和MS+2,4-D 0.5 mg/L+6-BA 1 mg/L的培养基上愈伤诱导率最高,分别为93.3%和96.2%。在不定芽再生培养过程中,6-BA和TDZ不能诱导不定芽发生,而ZT对不定芽的分化具有较好的诱导作用;"魁绿""佳绿"均在培养基MS+NAA 0.05 mg/L+ZT 3 mg/L上不定芽分化率最高,分别为72.0%、37.8%。不同品种及不同外植体的愈伤组织诱导率和不定芽再生率均存在差异,"魁绿"的不定芽分化率明显高于"佳绿";无芽茎段和叶片的愈伤组织诱导率均>90%;叶片的不定芽分化率最高,显著高于茎段和叶柄。再生苗在培养基1/2 MS+IBA 0.1 mg/L上的生根率为97%。 To establish the regeneration of "Kui Lü"and "Jia Lü",taking Actinidia arguta cultivar"Kui Lü"and"Jia Lü"stem segments,leaves and petioles as explants,the effects of explants types and plant growth regulator combination on callus induction and plantlet regeneration of different genotypes of Actinidia arguta were studied. The results show that the reactions of different genotypes to the culture conditions were different. The effect of 6-BA on callus induction of Actinidia arguta was better than that of KT. Neither 6-BA nor TDZ can induce A. arguta shoot differentiation,but ZT has a good effect on it. The best medium of "Kui Lü"leaf callus induction was MS+2,4-D 0. 5 mg/L+6-BA 2 mg/L,and the induction rate was 93. 3%; the best medium of "Jia Lü"leaf callus induction was MS+2,4-D 0. 5 mg/L+ 6-BA 1 mg/L with a 96. 2% callus induction rate. The differentiation rate was the highest when "Kui Lü"and "Jia Lü"callus was in the medium of MS + NAA0. 05 mg/L+ZT 3 mg/L,and the differentiation rate was 72. 0% and 37. 8%,respectively. Under the same culture conditions,the adventitious bud differentiation rate of"Kui Lü"was significantly higher than that of"Jia Lü".Different explants have different capabilities of callus induction and plant regeneration.Callus induction rates of stem without bud and leaf were above 90%;differentiation rate of leaf adventitious bud was the highest,significantly higher than that of stem and petiole.Compared with stem and petiole,leaf was more suitable to be used as explants of Actinidia arguta plant regeneration.The rooting rate of bud on 1/2 MS+IBA 0.1 mg/L was 97%.
