摘要
目的观察香烟烟雾提取物(CSE)对人支气管上皮细胞(16HBE)中人沉默信息调节因子6(SIRT6)、Caspase-9表达的影响。方法体外培养16HBE,加入不同浓度新鲜CSE诱导细胞,通过荧光定量聚合酶链反应(PCR)方法及Western blotting方法检测SIRT6及Caspase-9的表达量。比较(0%、5%、7.5%、10%)的CSE对16HBE中SIRT6、Caspase-9表达的影响。结果16HBE经过不同浓度(0%、5%、7.5%、10%)的CSE处理后,SIRT6mRNA表达差异有显著统计学意义(P〈0.01)。两两比较,各组间除5%CSE组和7.5%CSE组比较差异无统计学意义外(P〉O.05),其余各组两两比较差异均有显著统计学意义(P〈0.01)。Caspase-9mRNA表达差异有显著统计学意义(P〈O.01)。两两比较,各组间差异均有显著统计学意义(P〈0.01)。16HBE经过不同浓度(0%、5%、7.5%、10%)的CSE处理后。SIRT6蛋白、Caspase-9蛋白表达差异有显著统计学意义(P〈0.01)。两两比较,各组间差异均有显著统计学意义(P〈0.01或〈O.05)。结论CSE可引起16HBESIRT6、Caspase-9表达的改变。
Objective To observe the effect of cigarette smoke extract (CSE) on the expression of human silencing information regulator protein 6 (SIRT6) and Caspase-9 in human bronchial epithelial cells (16HBE). Methods In vitro culture of 16HBE, we added different concentrations of fresh CSE to induce cells. The expression of SIRT6 and Caspase-9 were detected by fluorescence quantitative polmerase chain reaction (PCR) and Western blotting. The effects of (0 %, 5 %, 7.5 %, 10 % )CSE on the expression of SIRT6 and Caspase-9 in 16HBE were compared. Results After 16HBE treatment with different concentrations (0 %, 5 %, 7.5 %, 10 % ) of CSE, the SIRT6 mRNA expression showed significant difference ( P d0.01). The differences between each two groups were statistically significant except for 5% CSE vs 7.5 % CSE ( P 〉0.05). Caspase-9 mRNA expression between groups showed significant difference ( P〈0.01).The differences between each two groups were statistically significant ( P〈0.01). After 16HBE treatment with different concentrations (0 %, 5 %, 7.5 %, 10 % ) of CSE, the SIRT6 protein expression and Caspase-9 protein showed significant difference between groups ( P〈0.01). The differences between each two groups were statistically significant ( P〈0.01 or〈0.05). Conclusions CSE can cause SIRT6, Caspase-9 of 16H13E expression changes.
