摘要 目的探讨HBV感染者精子长链非编码RNA(LncRNA)的表达情况及其意义。方法选择80例男性慢性HBV携带者及80例健康男性,应用基因芯片分析两组精子中差异表达的LncRNA。另收集HBV男性携带者及健康男性精液样本各5例,采用实时荧光PCR进行检测以验证LncRNA芯片数据的准确性。使用基因本体分析网站、京都基因与基因组百科全书数据库分别对差异表达LncRNA的靶基因进行功能富集、信号通路富集分析,采用GENECODE、UCSC数据库进行注释、鉴定和预测。结果与正常健康男性相比,HBV感染者精子中存在300个显著差异表达的LncRNA,其中214个表达上调,84个表达下调。差异表达LncRNA的靶基因在精氨酸生物合成、氨基酸生物合成、嗅觉转导、氨基酸(缬氨酸、亮氨酸和异亮氨酸)降解、p53信号转导途径、过氧化物酶体增殖物激活受体信号通路、肌动蛋白细胞骨架的调节和神经活性配体受体相互作用等调控过程中具有重要作用,这些通路主要涉及能量代谢、炎症反应、细胞分化与增殖、细胞凋亡等生物学过程。差异表达的LncRNA主要分布在1号、2号、5号、10号和16号染色体。结论 HBV感染者精子中存在显著差异表达的LncRNA,LncRNA可能在HBV感染导致的男性生育力下降发病机制中具有重要作用。 Objective To investigate the expression of long non-coding RNA( LncRNA) in sperm of males with HBV infection and its significance. Methods Eighty chronic HBV male carriers and 80 healthy males were enrolled,then gene chip technology was used to analyze differential expression of LncRNA in spermatozoa between the two groups. The sperm were collected from 5 chronic HBV male carriers and 5 healthy males,then the accuracy of LncRNA chip data was verified by real-time fluorescent PCR. Functional enrichment analysis and signaling pathway enrichment analysis were conducted using the website for Gene Ontology and the database of Kyoto Encyclopedia of Genes and Gnomes respectively. GENECODE and UCSC database were used for annotation,identification and prediction. Results Compared to the healthy males,300 LncRNAs with significantly differential expression were found in the sperm of males with HBV infection,including 214 up-regulated LncRNAs and 84 down-regulated LncRNAs. The target gene with LncRNAs with differential expression played important roles in many regulation processes including arginine biosynthesis,amino acid biosynthesis,olfactory transduction,degradation of amino acid( valine,leucine and isoleucin),p53 signal transduction pathway,peroxisome proliferator-activated receptor signaling pathway,regulation of actin cytoskeleton and neuroactive ligand-receptor interaction. Those pathways mainly involved biological processes such as energy metabolism,inflammatory response,cell differentiation and proliferation,and apoptosis. The LncRNAs with differential expression mainly distributed in chromosome 1,2,5,10 and 16. Conclusion LncRNAs with significantly differential expression exist in the sperm of males with HBV infection. LncRNA may play an important role in the pathogenesis of male fertility reduction caused by HBV infection.
