促肝细胞生长素对急性百草枯中毒大鼠肝组织的保护作用 被引量:2

摘要 目的探讨促肝细胞生长素(p HGF)对急性百草枯中毒大鼠肝组织的保护作用及其机制。方法将90只SD大鼠随机分为对照组、模型组和治疗组3组,每组30只。模型组、治疗组给予单次百草枯(25 mg/kg)腹腔注射建立模型,对照组腹腔注射等量生理盐水。治疗组建模后立即给予p HGF(50μg/kg)腹腔注射,其余两组腹腔注射等量生理盐水。于药物干预后第1、3、7天,各组各处死大鼠10只,检测血清ALT、AST、一氧化氮含量,观察肝组织病理学改变,检测肝组织B淋巴细胞瘤-2(Bcl-2)、Bcl-2相关X蛋白(Bax)的表达。结果在各时间点,模型组的ALT、AST水平均高于对照组及治疗组(P<0.05)。模型组各时间点的一氧化氮水平均高于对照组,第3天、第7天的一氧化氮水平均高于治疗组(均P<0.05)。在各时间点,模型组Bax、Bcl-2蛋白的表达水平均高于对照组,仅Bax的表达水平高于治疗组,而Bcl-2蛋白的表达水平低于治疗组(均P<0.05)。结论 p HGF可减轻急性百草枯中毒所致的肝组织损伤,这可能与p HGF抑制肝细胞的凋亡及降低血清一氧化氮水平有关。 Objective To investigate the protective effect of hepatocyte growth-promoting factor( p HGF) on liver in the rats with acute paraquat poisoning and its mechanism. Methods Ninety SD rats were randomly divided into control group,model group and treatment group,with 30 rats in each group. The model group and treatment group were intraperitoneally administered paraquat( 25 mg/kg) with single dose,and the control group was intraperitoneally administered normal saline of the same volume. After modeling,the treatment group was intraperitoneally administered p HGF( 50 μg/kg) immediately,and the other two groups were intraperitoneally administered normal saline of the same volume. On the 1 st,3 rd and 7 th day after administration,10 rats were sacrificed in each group. Then the contents of serum ALT,AST and nitric oxide( NO) were detected,the pathological changes of liver tissues were observed,and the expressions of liver tissue B-cell lymphoma-2( Bcl-2) and Bcl-2-associated X protein( Bax) were detected. Results At each time point,the levels of ALT and AST in the model group were higher than those in the control group and treatment group( P〈0. 05). The NO level at each time point was higher in the model group compared to the controls,and the NO levels on the 3 rd and 7 th day were higher in the model group than those in the treatment group( all P〈0. 05). At each time point,the expression levels of Bax and Bcl-2 protein in the model group were higher than those in the control group,and the expression level of Bax was higher but the expression level of Bcl-2 protein was lower in the model group compared to the levels in the treatment group( all P〈0. 05). Conclusion p HGF can alleviate the liver injury induced by acute paraquat poisoning,which is probably related to the inhibition of hepatocyte apoptosis and reduced NO level by p HGF.
