摘要
目的探讨老年人氧化型LDL(ox-LDL)与辅助性T细胞(Th)17及其细胞因子的相关性。方法选择2014年11月~2015年9月年龄≥60岁的老年人341例为老年组,年龄<60岁体检者107例为中年组。再依据ox-LDL水平三分位将老年人分为低水平组112例、中水平组115例和高水平组114例。检测血浆ox-LDL水平、外周血Th17细胞频率、白细胞介素(IL)17、TNF-α、IL-6及维甲酸相关孤独受体γt(RORγt)mRNA表达水平。结果与中年组比较,老年组Th17细胞频率、IL-17、ox-LDL及RORγt mRNA表达水平明显升高[(1.0±0.5)%vs(0.9±0.4)%,(29.8±12.3)ng/Lvs(26.6±12.6)ng/L,(379.3±101.6)μg/Lvs(302.8±94.0)μg/L,14.3±5.5vs 12.5±5.8,P<0.05,P<0.01],服用他汀类药物比例明显降低(11.4%vs 26.2%,P=0.002)。低、中、高水平各组TC、LDL-C、空腹血糖、Th17细胞频率、IL-17、IL-6、TNF-α及RORγt mRNA表达水平比较,差异有统计学意义(P<0.05,P<0.01)。多元线性逐步回归分析结果显示,ox-LDL是Th17细胞频率、IL-17、IL6、TNF-α及RORγt mRNA表达水平的独立影响因素(P<0.05,P<0.01)。结论老年人ox-LDL水平与外周血Th17细胞频率及其相关细胞因子密切相关。
Objective To study the association of ox-LDL with Th17 cells and their cytokines in the elderly.Methods Three hundred and forty-one≥60 years old subjects served as an old-aged group and 10760 years old subjects served as a middle-aged group.The subjects in old-aged group were divided into low ox-LDL group(n=112),moderate ox-LDL group(n=115)and high ox-LDL group(n=114).Their serum levels of ox-LDL,IL-17,TNF-α,IL-6,frequency of Th17 cells in peripheral blood and expression of RORγt mRNA in peripheral blood were measured.Results The frequency of Th17 cells,serum levels of IL-17,IL-6,TNF-αand expression level of RORγt mRNA were significantly higher while the administration rate of statins was significantly lower in old-aged group than in middle-aged group(P〈0.05,P〈0.01).The serum levels of TG,LDL-C,FBG,IL-17,IL-6,TNF-α,frequency of Th17 cells,expression level of RORγt mRNA were significantly different in 3 ox-LDL groups(P〈0.05,P〈0.01).The serum ox-LDL level was an independent influencing factor of the frequency of Th17 cells,serum levels of IL-17,IL-6,TNF-αand RORγt mRNA expression(P〈0.05).Conclusion The serum ox-LDL level is closely associated with the frequency of Th17 cells in peripherial blood and their cytokines in the elderly.
