Thermoascus aurantiacus产耐热木聚糖酶水解活性的培养条件优化 被引量:2

摘要 在前期研究的基础上,利用SPSS进行主成分分析,构建了催化水解玉米芯的综合水解能力指数(XCHI);然后以XCHI为自变量,水解玉米芯的还原糖得率为因变量进行回归分析,回归分析模型证明了综合水解能力XCHI与糖得率之间存在相关关系,以XCHI为响应值,正交实验优化得到T.aurantiacus CGMCC11334产Xyn水解玉米芯能力的最适培养基组成为:麸皮16 g/l、微晶纤维素7 g/l、牛肉浸膏4 g/l、玉米浆3 g/l、小麦蛋白胨4 g/l、KH_2PO_4 1.0 g/l、MgSO_4 2.0 g/l、CuSO_4 0.00 g/l、Fe-SO_4 0.05 g/l、VB1 0.15 g/l;在此条件下,Xyn、CMC和FPA的活性分别为135.12、1.95 U/ml和1.68 U/ml,XCHI为1 649.44。 Based on our previous results, the present study developed the comprehensive hydrolysis ability index (XCHI) by using principal component analysis. XCHI was proved to represent the hydroysis ability of xylanase by regressively analyzing the relation of XCHI and sugar yields with XCHI as in-dependent variable and reducing sugar yield as dependent variable. Using XCHI as response, the cul- ture compositions were optimized by using orthogonal experimental design as (g/l): wheat bran 16, mi-crocrystalline cellulose 7, beef extract 4, corn syrup 3, wheat peptone 4, KH2PO4 1.0, MgSO4 2.0, CuSO4 0.00, FeSO4 0.05, VB1 0.15. Under the optimal condition, the Xyn, CMC, FPA and XCHI were 135.12 U/ml, 1.95 U/ml, 1.68 U/ml and 1 649.44, respectively.
