摘要
介绍了葡聚糖接枝型琼脂糖分离介质的结构,随后对救火列队和静电耦合传质两种重要的传质机理进行了阐述,其中救火列队是一种特殊的表面扩散,依靠葡聚糖长链的摆动传递蛋白质,静电耦合传质表征金属盐离子对蛋白扩散的促进作用.最后介绍了近年来葡聚糖接枝型琼脂糖分离介质在层析领域的应用,其中在离子交换层析中的应用最为普遍,并已开发了多种商业化葡聚糖接枝型离子交换介质,在混合模式吸附层析和亲和层析领域的应用相对较少,可能与葡聚糖接枝层容易受盐及疏水基团等影响导致坍塌有关.因此,增强葡聚糖接枝层的结构稳定性对其应用具有重要意义.
In the present work,the structure of dextran-grafted agarose matrix was introduced firstly.Then important transport mechanisms including bucket brigade effect and electrostatic coupling effect were elaborated in detail.Bucket brigade effect is special surficial diffusion,and protein is transmitted through the long chain of dextran.Electrostatic coupling effect characterizes promotion of protein diffusion in the presence of metal salt ions.Finally,the applications of dextran-grafted agarose resins in chromatography were reviewed.To date,dextran-grafted agarose resins were mainly applied in ion exchange chromatography,and a variety of commercial dextran-grafted ion exchange media have been developed.While there were fewer applications in mixed mode chromatography and affinity chromatography,since dextran-grafted layer is susceptible to collapse influenced by salt and hydrophobic groups.Therefore,enhancing the structural stability of dextran-grafted layer is of great significance to its application.
