摘要
目的:探讨斑秃患者外周血单个核细胞(PBMCs)中转录因子叉状头螺旋转录因子(FOXP)3蛋白的表达水平。方法 :将52例斑秃患者分为活动期组18例,稳定期组34例;局限型斑秃组41例,重型斑秃组11例。选取50例健康人作为正常对照组。采用蛋白质印迹法(WB)检测2组PBMCs中FOXP3蛋白表达水平,并进行相关性分析。结果:正常对照组PBMCs中FOXP3蛋白表达均值为1.33±0.66,斑秃患者活动期组为0.45±0.34,稳定期组为0.80±0.42,3组间比较差异有统计学意义(F=20.89,P<0.05);活动期组和稳定期组蛋白表达水平均低于正常对照组(P<0.05),斑秃活动期组低于稳定期组(P<0.05)。重型斑秃组FOXP3蛋白表达均值为0.41±0.26,局限型斑秃组为0.75±0.43,与正常对照组比较,3组蛋白表达差异有统计学意义(F=19.77,P<0.05);且重型斑秃组和局限型斑秃组均低于正常对照组(P<0.05),重型斑秃组低于局限型斑秃组(P<0.05)。斑秃患者PBMCs中FOXP3蛋白表达水平与斑秃病情的严重程度(SALT)呈负相关(r=-0.33,P<0.05)。结论:斑秃患者PBMCs中转录因子FOXP3蛋白的表达水平降低可能与斑秃的细胞免疫失调有关。
Objective: To investigate the expression of transcription factor FOXP3 in peripheral blood mononuclear cells (PBMCs) from patients with alopecia areata(AA). Methods: Western blot was used to measure expression levels of FOXP3 protein in PBMCs from 52 AA patients and 50 healthy controls. Results: Expression levels of FOXP3 in the active and stable AA patients, and normal controls were 0.45±0.34, 0.83±0.40 and 1.33±0.66, respectively(F:20.89,P〈0.05). FOXP3 levels in active AA were lower than that in stable AA(P〈O.05). FOXP3 levels in either active or stable AA were lower than that in normal controls(P〈0.05). Moreover, expression levels of FOXP3 in localized and severe AA differed significantly(0.75±0.43 in localized versus 0.41±0.26 in severe, P〈0.05). Furthermore, FOXP3 levels in both localized and severe AA differed significantly from normal controls(P〈0.05). Finally, expression levels of FOXP3 negatively correlated with SALT in patients with AA (r=-0.33, P〈0.05). Conclusions: Decreased expression levels of the transcription factor FOXP3 protein in PBMCs may be related to dysregulation of cellular immunity in AA.
