PHB在结肠癌中的表达及临床病理关系研究

The study of correlationship between the expression of PHB in colon cancer and its clinicopathological charasteristics

目的探究PHB蛋白在结肠癌组织及癌旁组织中的表达情况,分析其与结肠癌病理分级、临床分期和浸润程度等临床病理特征的关系,并探讨PHB在结肠癌发生、发展中的作用及临床意义。方法收集结肠癌患者手术切除的癌组织及对应的距离病变组织5cm以上的癌旁正常组织各30例。利用HE染色核对组织病理类型,并通过免疫组织化学染色技术,检测结肠癌组织及相应癌旁正常的结肠组织中PHB的表达水平。分析PHB的表达水平与患者年龄、性别、病理分级、临床分期及浸润程度之间的关系。结果 HE染色结果显示收集的30例结肠癌组织病理类型均为腺癌。免疫组织化学染色显示,结肠癌组织细胞中PHB阳性表达呈黄色或棕黄色颗粒,主要定位于胞质。在30例正常的结肠组织中,PHB呈阳性表达的有10例,呈阴性表达的有20例,阳性率为33.3%;在30例结肠癌组织中,PHB呈阳性表达的有20例,呈阴性表达的有10例,阳性率为66.7%。PHB的表达水平与患者临床分期有关,随着患者临床分期增加而逐渐增高(P<0.05),与患者的性别、年龄、病理分级及浸润程度无明显相关性(均P>0.05)。结论结肠癌患者癌组织中PHB的阳性表达率增高,且PHB的高表达现象与患者临床分期相关,提示PHB在结肠癌的发生与发展中发挥一定的作用。

Objective To investigate the expression levels of PHB in colon cancer tissues and corresponding normal colon tissues,and to analyze their correlation with the chinicopathological characteristics,including pathological grade,clinical stage and infiltration degree. At the same time,the role of PHB was explored in the development and progression of colon cancer and its clinical significance. Methods Thirty colon cancer tissues from surgical resection and their adjacent normal tissues at more than 5 cm away from lesioned tissues were collected from the First People＇s Hospital of Pingdingshan from 2015 to 2017. Histopathological types were examined by HE staining. Immunohistochemical staining method was used to assess the expression level of PHB in colon carcinoma and paired normal colonic tissues. The correlation between the staining expression and clinicopathological features,including gender,age,clinical stage,pathological grade and infiltration degree,were examined. Results HE staining showed that 30 cases of colorectal cancer were classified as adenocarcinoma. Immunohistochemistry showed positive expression of PHB as yellow or brown particles,and mainly located in the cytoplasm of colonic epithelial cells. Among 30 samples of adjacent normal tissues,PHB-positive expression was revealed in 10 cases. Among 30 cases of colon cancer tissues,PHB-positive expression was revealed in 20 cases. The PHB positive rate of colon cancer and normal colonic tissue were 66. 7% and 33. 3%,respectively,with significant difference between them（ P〈0. 05）. PHB-positive expression was significantly associated with the clinical stage（ P〈0. 05）,but not with gender,age,pathological grade or infiltration degree（ P〉0. 05）. Conclusion In colon cancer,PHB is significantly up-regulated and significantly associated with clinical stage. PHB may play an important role in the occurrence and development of colon cancer.
